摘  要:目的探讨S100A7在结直肠癌组织中表达及沉默结直肠癌HCT116细胞中S100A7基因表达对细胞增殖和侵袭的影响。方法选取结直肠癌患者136例,截止2018年6月30日,获得随访125例(91.91%),用Kaplan-Meier和Log-Rank法进行生存分析,Cox比例风险回归模型分析患者预后影响因素。免疫组化法检测结直肠癌和癌旁组织中S100A7蛋白表达。人结直肠癌HCT116细胞分为S100A7干扰序列组(转染S100A7 siRNA序列)、阴性对照组(转染阴性对照序列)和空白组(不作任何处理),实时荧光定量PCR检测细胞中S100A7基因表达,CCK-8法检测细胞增殖,Transwell法检测细胞侵袭能力,蛋白质印迹检测细胞中S100A7、E-cadherin和Vimentin蛋白表达。结果结直肠癌组织中S100A7蛋白阳性表达率高于癌旁组织中(77.9%vs 33.1%,χ^2=55.394,P<0.001);Kaplan-Meier生存分析结果显示,S100A7蛋白阳性表达组患者平均生存时间和累积生存率均低于S100A7蛋白阴性表达组(χ^2=9.935,P=0.002);Cox比例风险回归模型显示,TNM分期、淋巴结转移和S100A7蛋白表达是影响患者预后的风险因素(HR=2.181,2.121,2.228,均P<0.05)。与阴性对照组和空白组比较,S100A7干扰序列组细胞中S100A7 mRNA和蛋白相对表达量降低(P<0.001),24,48,72,96 h时OD值降低(均P<0.05),侵袭细胞数减少(P<0.05),细胞中Vimentin蛋白相对表达量降低(P<0.001),而E-cadherin蛋白相对表达量则升高(P<0.001)。结论S100A7蛋白在结直肠癌组织中呈高表达,沉默结直肠癌细胞中S100A7基因表达可抑制细胞增殖及侵袭,其机制可能是沉默S100A7基因表达抑制了上皮-间质转化过程。Objective To investigate the expression of S100A7 in colorectal cancer tissue,and explore the effect of silencing the expression of S100A7 gene on proliferation and invasion of colorectal cancer HCT116 cells.Methods A total of 136 patients with colorectal cancer were selected,and followed-up after surgery until June 30,2018.A total of 125 patients were followed up,and the follow-up rate was 91.91%.Kaplan-Meier and Log-Rank methods were used for survival analysis.Cox proportional hazards regression model was used to analyze the factors affecting patient prognosis.The expression of S100A7 protein in colorectal cancer and adjacent tissues was detected by immunohistochemistry.The human colorectal cancer HCT116 cells were cultured and divided into S100A7 interference sequence group(transfected with S100A7 siRNA sequence),negative control group(transfected with negative control sequence)and blank group(no any treatment).The expression of S100A7 gene in cells was detected by real-time fluorescent quantitative PCR.Cell proliferation was detected by CCK-8 assay.Cell invasion was detected by Transwell assay.The expression of S100A7,E-cadherin and Vimentin proteins was detected by Western blot.Results The positive expression rate of S100A7 protein in colorectal cancer tissues was higher than that of adjacent tissues(77.9%vs 33.1%,χ^2=55.394,P<0.001).Kaplan-Meier survival analysis showed that the average survival time and cumulative survival rate in S100A7 protein positive expression group were lower than those in S100A7 protein negative expression group(χ^2=9.935,P=0.002).Cox proportional hazards regression model showed that TNM stage,lymph node metastasis and S100A7 protein expression were risk factors for prognosis(HR=2.181,2.121,2.228,all P<0.05).Compared with negative control group and blank group,the relative expression levels of S100A7 mRNA and protein in S100A7 interference sequence group were decreased(P<0.001),the OD values at 24 h,48 h,72 h and 96 h were decreased(all P<0.05),the number of invasive cells was dec
