摘  要:以毛泡桐(Paulownia tomentosa)病苗(Pto)和健康苗(PtoI)为试验材料,利用高通量测序技术对染病前后毛泡桐的叶绿体全基因组进行测序,分析比较基因组结构。研究结果表明,毛泡桐病苗和健康苗的叶绿体基因组由反向重复区(IRs)、长单拷贝序列区(LSC)和短单拷贝序列区(SSC)等部分组成。其中,叶绿体基因组全长分别为154 700 bp(Pto)和154 646 bp(PtoI),IRs的长度为25 779 bp,LSC和SSC长度分别为85 356、17 786 bp(Pto)和85 356、17 732 bp(PtoI)。生物信息学分析表明,在Pto中共搜索到简单重复位点(SSR)64个,单核苷酸、二核苷酸、三核苷酸、四核苷酸和五核苷酸的数量分别为49、5、2、7、1个;PtoI中共有64个SSR位点,单核苷酸个数为49个,并未发现六核苷酸。大多数位点都富含A-T,具有碱基偏好性。The high-throughput sequencing technology was utilized to analyze and compare the structure of chloroplast genome using healthy and infected seedlings,with PaWB(PtoI),of Paulownia tomentosa(Pto).The result showed that the structure of chloroplast genome was composed of one large single-copy(LSC)region,one small single-copy(SSC)region and a couple of inverted repeats(IRa and IRb).Among them the size of chloroplast genome of Pto and PtoI was 154 700 bp and 154 646 bp,respectively.Besides,the length of the IRs was 25 779 bp,and the length of LSC and SSC was 85 356 bp,17 786 bp(Pto)and 85 356 bp,17 732 bp(PtoI),respectively.64 simple sequence repeats(SSR)loci were found in Pto chloroplast genome through bioinformatics analysis.The number of mono-,di-,tri-,tetra-and pentanucleotide repeat motifs was 49,5,2,7 and 1,respectively.64 SSRs loci were obtained in PtoI,mononucleotide repeat motif were 49.No hexa-nucleotide repeat was found.Most SSR loci were rich in A-T and had a base preference.
