摘  要:目的观察硼酸钠对人肝癌HepG2细胞Hedgehog信号通路蛋白表达的影响。方法将HepG2细胞随机分为两组,实验组用4.0 mmol/L硼酸钠处理24 h,对照组不做处理;收集细胞,采用实时荧光定量PCR(qRT-PCR)法检测细胞中Hedgehog信号通路蛋白Gli2、Ptch1、Cyclin D1 mRNA,Western blotting法检测细胞中Hedgehog信号通路蛋白Gli2、Ptch1、Cyclin D1。结果实验组细胞中Gli2、Ptch1、Cyclin D1 mRNA相对表达量分别为0.58±0.05、0.64±0.08、0.35±0.03,相应的蛋白相对表达量分别1.14±0.03、0.93±0.02、1.86±0.02,均低于对照组,差异有统计学意义(P均<0.05)。结论硼酸钠可下调人肝癌HepG2细胞中Hedgehog信号通路蛋白的表达,为新型抗肿瘤药物的研发及作用靶点提供了依据。Objective To observe the effect of borax on Hedgehog signaling pathway protein expression of human hepatocellular carcinoma HepG2 cells.Methods HepG2 cells were randomly divided into two groups.The experimental group was treated with 4.0 mmol/L borax for 24 h,and the control group was not treated.The cells were collected and the Hedgehog signaling pathway proteins Gli2,Ptch1 and Cyclin D1 were detected by real-time quantitative PCR(qRT-PCR).The Hedgehog signaling pathway proteins Gli2,Ptch1 and Cyclin D1 were detected by Western blotting.Results The relative mRNA expression levels of Gli2,Ptch1 and Cyclin D1 were 0.58±0.05,0.64±0.08,and 0.35±0.03,respectively,and the corresponding protein relative expression levels were 1.14±0.03,0.93±0.02,and 1.86±0.02,respectively,which were lower than those of the control group,with statistically significant differences(all P<0.05).Conclusion Borax may down-regulate the expression of Hedgehog signaling pathway protein in human hepatocellular carcinoma HepG2 cells,which provides a basis for the development and target of new anti-tumor drugs.
