摘 要:目的:观察复方苦参注射液对放射性肺炎大鼠肺组织Notch1及Jagged1信号通路表达的影响。方法:将40只SD大鼠随机分为正常组、模型组、醋酸泼尼松龙组、复方苦参注射液组,直线加速器对除正常组外3组大鼠全胸单次照射16Gy。照射后各组予相应的药物灌胃。4周后处死大鼠,计算肺系数,取大鼠肺组织进行HE染色、Masson染色,Notch1免疫组化染色,RT-PCR、Western blot测定Notch1、Jagged1 mRNA及相关蛋白的表达水平。结果:与正常组比较,模型组大鼠肺系数显著增大(P<0.05),Notch1、Jagged1 mRNA表达量显著增高(P<0.05),Notch1、NICD、Jagged1蛋白表达量显著提高(P<0.05);与模型组比较,复方苦参注射液组和醋酸泼尼松龙组可显著降低肺系数和Notch1、Jagged1 mRNA表达量(P<0.05),显著下调Notch1、NICD、Jagged1相关蛋白表达(P<0.05)。结论:复方苦参注射液能明显下调放射性肺炎大鼠肺系数和Notch1、Jagged1,可能是其防治放射性肺炎发生发展的机制之一。Objective: To observe the effects of Compound Kushen Injection on the expression of Notch1 and Jagged1 signaling pathway in lung tissue of rats with radiation pneumonitis. Methods: Forty male and female SD rats were randomly divided into normal group, model group, prednisolone acetate group and Compound Kushen Injection group. The dose was 16 Gy. After irradiation, each group was given corresponding drugs by gavage. Four weeks later, rats were sacrificed and their lung coefficients were calculated. The lung tissues were harvested by HE staining, Masson staining and Notch1 immunohistochemical staining. The expression levels of Notch1, Jagged1 mRNA and related proteins were measured by RT-PCR and Western blot. Results: Compared with the normal group, the lung coefficient of the model group was significantly higher(P<0.05), the expression of Notch1 and Jagged1 mRNA were significantly increased(P<0.05), the expression of Notch1, NICD and Jagged1 related proteins were significantly up-regulated(P<0.05). Compared with the model group, Compound Kushen Injection group and prednisolone acetate group significantly reduced the lung coefficient and Notch1 and Jagged1 mRNA expression(P<0.05), and significantly down-regulate the expression of Notch1, NICD and Jagged1 related proteins(P<0.05). Conclusion: Compound Kushen Injection can significantly reduce Notch1 and Jagged1 in rats with radiation pneumonia, which may be one of the mechanisms of preventing and treating the occurrence and development of radiation pneumonia.
