Folin-Ciocalteu比色法测定昭通青花椒总酚含量

摘　　要：用60%乙醇溶液于25℃超声提取昭通青花椒的酚性成分。以没食子酸为标准品,采用Folin-Ciocalteu比色法,在765 nm处测得标准曲线为y=0.0873x+0.0095(r=0.9991),该方法在0~5μg/mL范围内总酚浓度与吸光度线性关系良好。经测定,昭通七县区青花椒总酚含量范围为52.57~62.07 mg/g,平均总酚含量为56.04 mg/g,回收率为100.1%,相对标准偏差(RSD)为0.39%(n=6),此方法操作简单,测定结果准确可靠。该研究结果可为昭通青花椒的开发利用提供试验参考。The phenol content in Zhaotong Zanthoxylum schinifolium is ultrasonically extracted in 60%ethanol solution at 25℃.Using gallic acid as the standard sample,Folin-Ciocalteu colorimetry is used.The standard curve at 765 nm is y=0.0873x+0.0095(r=0.9991).The linear relationship between total phenol concentration and absorbance is good in the range of 0~5μg/mL.The total phenol content of Zanthoxylum schinifolium in seven areas of Zhaotong is in the range of 52.57~62.07 mg/g.The average total phenol content is 56.04 mg/g.The recovery rate is 100.1%.The relative standard deviation(RSD)is 0.39%(n=6).The method is simple and accurate.The results can provide experimental reference for the development and utilization of Zanthoxylum schinifolium in Zhaotong.
