摘  要:通过富集培养后从鱼体表分离得到了有活性的蛭弧菌,双层平板培养后能出现明显的透明状噬菌斑;用银染法染色后观察到蛭弧菌呈逗点状,同时观察到蛭弧菌的连续运动状态;通过16S rRNA基因序列的同源性分析结果表明,分离到的鱼体表蛭弧菌与大多数典型蛭弧菌的同源性均较高,最高达99%,可以分类到Bdellovibrio属。该实验内容与生活贴近且涉及实验技术广泛,有助于激发学生的实验热情,促进创新型人才的培养,也为今后在微生物学实验教学中开设综合创新实验奠定了基础。An active strain of Bdellovibrio bacteriovorus was isolated from the body surface of fish after enrichment culture,and the Bdellovibrio bacteriovorus can be observed by culturing in double plates.By silver nitrate staining method,the Bdellovibrio bacteriovorus was stained.The Bdellovibrio bacteriovorus morphology and process of invading E.coli could be clearly observed and recorded using the digital imaging system of phase contrast microscope.The bacterial 16S rRNA gene sequence homology analysis showed that it was most close to most typical Bdellovibrio bacteriovorus strains and could be classified into Bdellovibrio.The experiments based on daily life and involving extensive experimental techniques,can not only stimulate enthusiasm on microbial experiments,but also promote the cultivation of innovative talents,and laid the foundation for the comprehensively innovative experiment in microbiology experimental teaching.
