摘  要:目的研究青少年唾液的群落结构,预测物种的功能及代谢信息。方法本研究采集非活动性龋的青少年及成年人的非刺激性唾液,利用16S rRNA基因高通量测序进行物种组成、多样性及差异物种等分析。利用PICRUSt对16S rRNA基因进行功能预测。结果青少年唾液样本的物种α多样性比成年人低。青少年组放线菌属OTU55的相对丰度高于成年组,而卟啉单胞菌属OTU9,牙髓卟啉单胞菌OTU51,普氏菌属OTU315,中间普氏菌OTU 28,密螺旋体属OTU74,Filifactor OTU59的相对丰度成年组高于青少年组。在KEGG第三级通路上,半胱氨酸和甲硫氨酸代谢在成年组更高,且与卟啉单胞菌属OTU9,牙髓卟啉单胞菌OTU51,中间普氏菌OTU 28,密螺旋体属OTU74的相对丰度呈正相关关系。结论青少年唾液的群落结构及功能与成年人有差异,该研究为开展该年龄段的口腔微生物研究提供参考。Objective To investigate the salivary microbial communities of teenagers.Methods In this study,16 teenagers and 13 adults without active dental caries were recruited.The microbial communities'structure was analyzed using 16S rRNA gene high-throughput sequencing,including microbial composition,diversity and OTUs with different relative abundances.Furthermore,a method called“PICRUSt”was used to make functional predictions based on 16S rRNA gene.Results Theαdiversity of teenagers was lower than that of adults.The relative abundance of Actinomyces OTU55 was higher in saliva from teenagers,while Porphyromonas OTU9,Porphyromonasendodontalis OTU51,Prevotella OTU315,Prevotellaintermedia OTU 28,Treponema OTU74,Filifactor OTU59 had higher relative abundance in saliva samples of adults.At KEGG level 3,Cysteine and Methionine Metabolism increased in adults'saliva samples,with positive correlations between Porphyromonas OTU9,P.endodontalis OTU51,P.intermedia OTU 28and Treponema OTU74.Conclusion The salivary microbial community and functions of teenagers and adults were different in some aspects.
