摘 要:目的 探讨发现于炎症区域1(FIZZ1)对大鼠主动脉平滑肌细胞(RASMC)的收缩作用机制。方法 采用贴壁法培养RASMC,随机分为正常对照组,阳性对照组(血管紧张素Ⅱ终浓度1×10^-7 mol/L),FIZZ1刺激组、2倍FIZZ1刺激组和4倍FIZZ1刺激组(分别用1×10^-8 mol/L、2×10^-8 mol/L和4×10^-8 mol/L FIZZ1),调钙蛋白(CaM)抑制剂组和肌球蛋白轻链激酶(MLCK)抑制剂组(分别采用FIZZ1+CaM抑制剂W-7或MLCK抑制剂ML-7),各组分别刺激RASMC 48h。用RT-PCR测定各组CaM和MLCK mRNA表达。结果 4倍FIZZ1刺激组CaM mRNA显著高于正常对照组和阳性对照组,差异有统计学意义(1.22±0.31 vs 0.28±0.06和0.60±0.10,P<0.05)、且MLCK mRNA表达水平显著高于正常对照组和阳性对照组,差异有统计学意义[(7.92±2.60)×10^-4 vs (2.01±1.08)×10^-4和(4.80±1.21)×10^-4,P<0.05]。CaM抑制剂组和MLCK抑制剂组较3个FIZZ1刺激组CaM和MLCK mRNA表达明显降低,差异有统计学意义(P<0.05)。结论FIZZ1可能通过Ca2+-CaMMLCK途径引起RASMC收缩。Objective To study the contraction mechanism of rat aortil smooth muscle cells(RASMC)in rat aorta wall by found in inflammatory zone 1(FIZZ1).Methods The adherently cultured RASMC were divided into normal control group,positive control group(1×10^-7 mol/L AngⅡ),FIZZ1 stimulation group(1×10^-8 mol/L FIZZ1),2-fold FIZZ1 stimulation group(2×10^-8 mol/L FIZZ1),4-fold FIZZ1 stimulation group(4×10^-8 mol/L FIZZ1),CaM inhibitor group and MLCK inhibitor group.The expressions of CaM and MLCK mRNA in different groups were detected by RT-PCR after the RASMC were stimulated for 48 h.Results The expression levels of CaM and MLCK mRNA were significantly higher in 4-fold FIZZ1 stimulation group than in normal control group and positive control group(1.22±0.31 vs 0.28±0.06 and 0.60±0.10,P<0.05;7.92×10^-4±2.60×10^-4 vs 2.01×10^-4±1.08×10^-4 and 4.80×10^-4±1.21×10^-4,P<0.05)while those of CaM and MLCK mRNA were significantly lower in CaM inhibitor group and MLCK inhibitor group than in FIZZ1 stimulation group,2-fold FIZZ1 stimulation group and 4-fold FIZZ1 stimulation group(P<0.05).Conclusion FIZZ1 can induce the contraction of RASMC in rat aortic wall through the Ca2+-CaM-MLCK pathway.
