摘  要:目的:探讨IL-17在人牙周膜成纤维细胞(hPDLFs)中的表达对牙周炎伴口腔扁平苔藓患者中的作用及意义。方法:采用ELISA方法检测43例健康志愿者(对照组)和43例牙周炎伴口腔扁平苔藓患者(研究组)血清中IL-17蛋白表达水平及IL-17在研究组唾液中的表达水平;检测研究组牙周指标。CCK8法检测IL-17对hPDLFs细胞增殖能力的影响;qRT-PCR和ELISA方法分别检测IL-17对hPDLFs中MMP1,MMP2,MMP3和MMP9的mRNA及蛋白质表达水平影响。通过Western blot及ELISA检测IL-17对hPDLFs细胞中NF-kB P65蛋白磷酸化的影响及MMP1,MMP2,MMP3和MMP9的蛋白表达变化。结果:研究组血清IL-17水平明显高于对照组(P<0.05),唾液中IL-17水平与牙周破坏程度呈正相关。IL-17可促进hPDLFs中NF-kB P65蛋白磷酸化,并促进MMP-1,MMP-2,MMP-3和MMP-9 Mrna及蛋白的表达。结论:IL-17可通过激活NF-kB信号通路促进hPDLFs中MMP-1,MMP-2,MMP-3和MMP-9的分泌。Objective: To explore the effect and significance of IL-17 expression in human periodontal ligament fibroblasts(hPDLFs), serum and saliva of periodontitis patients with oral lichen planus(OLP). Methods: The expression level of IL-17 protein in the serum of 43 healthy volunteers(control group) and 43 patients with periodontitis accompanied by oral lichen planus(OLP, case group) and the expression level of IL-17 in case group was measured by ELISA. Clinical periodontal indicators were detected in case group. The effect of IL-17 on proliferation of hPDLFs was detected by CCK8. The effect of IL-17 on mRNA expression level and protein secretion of MMP-1,MMP-2,MMP-3 and MMP-9 in hPDLFs was detected by QRT-PCR and ELISA. The effect of IL-17 on the phosphorylation of NF-KB P65 protein in hPDLFs and the protein expression changes of MMP-1,MMP-2,MMP-3 and MMP-9 were detected by ELISA. Results: The expression level of IL-17 in the serum of case group was higher than that of control group, and the expression level of IL-17 in saliva was positively correlated with the degree of periodontal destruction. IL-17 promoted the phosphorylation of NF-KB P65 protein in hPDLFs, and promoted the mRNA expression and protein secretion of MMP-1, MMP-2, MMP-3 and MMP-9. Conclusion: IL-17 may activate NF-kB signaling pathway and promote the secretion of MMP-1, MMP-2, MMP-3 and MMP-9 in hPDLFs in patients with periodontitis and OLP.
