摘 要:目的探讨类胰岛素生长因子I受体(insulin-like growth factor 1 receptor,IGF1R)反义长链非编码RNA-IRAIN与喉癌发生的相关性。方法选取37例喉癌及癌旁组织和6例正常咽部黏膜,检测IRAIN等位基因印记表达,qPCR研究IRAIN和IGF1R在喉癌组织及癌旁组织中的表达。统计学分析IRAIN和IGF1R在喉癌组织、癌旁组织中的表达关系及与喉癌患者临床特征之间的关系。结果将gDNA测序IRAIN SNP位点(rs8034564)为杂合子的患者选出后行cDNA测序,IRAIN在喉癌组织、癌旁组织和正常咽部黏膜中均呈双等位基因表达;IGF1R在喉癌组织中较癌旁组织呈高表达(P<0.05),IRAIN在喉癌组织的表达低于癌旁组织(P<0.05),喉癌患者的年龄、TNM分期等临床特征与IRAIN、IGF1R的表达无统计学意义(P均>0.05)。在喉癌组织癌旁组织存在等位基因的不平衡表达(allelic expression imbalance,AEI),正常组织中不存在AEI。结论 IRAIN等位基因在正常咽部黏膜、喉癌组织及癌旁组织中呈非印记表达。IGF1R在喉癌组织中的表达高于癌旁组织,可能与喉癌组织的发生发展有关。在喉癌组织中IRAIN表达低于癌旁组织,IRAIN可能作为抑癌因素参与喉癌的发生。在喉癌与癌旁组织存在AEI,可能作为喉癌诊断指标。OBJECTIVE To discuss the relationship between long non-coding RNA of IGF1R-IRAIN and laryngeal carcinoma. METHODS Six samples of normal soft palate tissue, carcinoma tissues and para-carcinoma tissues from 37 patients with surgical treatment of laryngeal cancer were collected. There was an informative single nucleotide polymorphism(SNP) rs8034564 to distinguish the two parental alleles of IRAIN. We extracted DNA and RNA from tissues and sequenced IRAIN allelic gene by PCR. Quantitative PCR was used to study the expression of IRAIN and IGF1R in laryngeal carcinoma and paracancerous tissues. RESULTS IRAIN was diallelic expression in normal soft palate tissues and adjacent laryngeal specimens, as well as in laryngeal cancer tissues. The expression level of IGF1R is highly expressed in laryngeal cancer tissues compared with paracancerous tissues(P<0.05). The expression of IRAIN in laryngeal cancer tissues was lower than that in paracancerous tissues(P<0.05). We found that AEI existed in laryngeal carcinoma and paracarcinoma, without in normal tissues. CONCLUSION IRAIN allele was non-imprinted in laryngeal carcinoma. IRAIN may be a potential tumor suppressor in laryngeal carcinoma. The AEI detection of IRAIN may be an early diagnosis marker of laryngeal carcinoma.
