摘 要:目的:研究甘薯膳食纤维(SPDF)对非酒精性脂肪性肝炎(NASH)小鼠的防护作用及相关机制。方法:30只雄性C57BL/6小鼠适应性饲养2周,随机分为正常对照组(C组)10只、模型组(M组)10只、甘薯膳食纤维组(SPDF组)10只。高脂饮食联合聚糖硫酸钠盐12周建立NASH模型。SPDF组小鼠在造模同时予以SPDF (0.7 g/kg)悬浊液灌胃干预。每周称取小鼠体重,实验结束后检测血清总胆固醇(TC)、甘油三酯(TG)、内毒素(LPS)水平;HE染色观察小鼠肝组织病理学变化, Western blotting方法测定肠道组织ZO-1、Occludin、Claudin-1的蛋白表达。结果:M组与C组小鼠相比,体重、肝重、肝指数显著升高;SPDF组与M组小鼠比较,体重、肝重、肝指数下降明显,且差异有统计学意义(P<0.01)。HE染色显示M组小鼠肝脏有明显脂质沉积以及炎细胞浸润,SPDF组小鼠肝组织脂肪变性程度明显减轻,脂滴数量减少,炎性细胞浸润减少。M组小鼠血清TC、TG、LPS水平明显高于C组;SPDF组小鼠血清TC、TG、LPS水平明显降低,差异有统计学意义(P<0.05)。较之C组,M组小鼠的肠道紧密连接蛋白ZO-1、Occludin、Claudin-1表达下降明显,而SPDF组的表达上调。结论:SPDF有可能通过肠肝轴上调肠道紧密连接蛋白ZO-1、Occludin、Claudin-1的表达,降低肠道通透性和血清内毒素LPS的水平,对NASH小鼠起防护作用。Objective:To study the effects and mechanisms of sweet potato dietary fiber(SPDF) on nonalcoholic steatohepatitis(NASH) mice. Methods:Thirty male C57 BL-6 mice were housed for 2 weeks. They were randomly divided into normal control group(group C, N=10), model group(group M), and sweet potato dietary fiber group(SPDF group). The high-fat diet was combined with DSS and the NASH model was established for 12 weeks. The SPDF group was intervened with SPDF 0.7 g/kg while modeling. The body weight of the mice was weighed weekly. After the experiment, serum total cholesterol(TC), triglyceride(TG) and endotoxin(LPS) levels were detected. HE staining was used to observe the pathological changes of liver tissue in mice. Western blotting was used to determine the intestinal protein expression of ZO-1, Occludin, and Claudin-1. Results:Compared with group C, body weight, liver weight and liver index were significantly higher in group M. Compared with group M, body weight, liver weight and liver index decreased significantly, and there was statistical difference(P<0.01). HE staining of liver tissue showed obvious lipid deposition and inflammatory cell infiltration in M group. The degree of steatosis in liver tissue of SPDF group was significantly reduced, the number of lipid droplets decreased as well as inflammatory cell infiltration. The levels of TC, TG and LPS of M group were significantly higher than those of normal control group;the levels of serum TC, TG and LPS in SPDF group were significantly lower(P<0.05). compared with group C, the intestinal expression of tight junction proteins ZO-1, Occludin and Claudin-1 decreased significantly in model group, while the expression of SPDF group was up-regulated.Conclusion:SPDF can up-regulate the expression of tight junction proteins ZO-1, Occludin and Claudin-1 in the intestinal trunk, reduce intestinal permeability and serum endotoxin LPS levels, and then protect NASH.
