摘  要:目的:观察蓝莓对肝纤维化模型中抗纤维化转录因子Krüppel样因子11(KLF11)的影响,探讨其可能的机制。方法:40只健康同周龄SD大鼠被随机分为正常对照组、蓝莓对照组、模型组和蓝莓预防组,每组10只,使用四氯化碳制作肝纤维化模型。测定各组大鼠血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)、Ⅳ型胶原、Ⅲ型前胶原(PCⅢ)、透明质酸(HA)和层粘连蛋白水平;Masson和HE染色对肝组织进行病理学检查;Western blot和免疫组织化学染色检测肝组织中KLF11的蛋白表达,Western blot检测肝组织中α-平滑肌肌动蛋白(α-SMA)的蛋白表达;real-time PCR检测肝组织中KLF11和Ⅰ型胶原(ColⅠ)的mRNA表达情况。结果:与正常对照组比较,模型组KLF11蛋白表达显著降低(P<0.01),而α-SMA蛋白表达显著升高(P<0.01);与模型组比较,蓝莓预防组KLF11的mRNA表达显著升高(P<0.01)而ColⅠ的mRNA表达显著降低(P<0.01);与模型组比较,其它各组大鼠血清ALT、AST、HA和PCⅢ水平均显著降低(P<0.01),蓝莓预防组大鼠肝纤维化分级显著降低(P<0.01)。结论:蓝莓对四氯化碳所致大鼠肝纤维化有一定的预防作用,其机制可能与其影响KLF11表达有关。AIM:To observe the effects of blueberry on anti-fibrosis transcription factor Krüppel-like factor 11(KLF11)in rat model of hepatic fibrosis.METHODS:Healthy SD rats(n=40)of the same age were randomly divided into normal control group,blueberry control group,model group and blueberry prevention group with 10 rats in each group.Liver fibrosis model was established by injection of carbon tetrachloride.The levels of serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),collagen typeⅣ,procollagen typeⅢ(PCⅢ),hyaluronic acid(HA)and laminin were detected.The pathological changes of liver tissues were observed after Masson and HE staining.The protein expression of KLF11 in the liver tissue was detected by Western blot and immunohistochemical staining,while the protein expression ofα-smooth muscle actin(α-SMA)in the liver tissues was detected by Western blot.The mRNA expression of KLF11 and collagen typeⅠ(ColⅠ)in the liver tissues was detected by real-time PCR.RESULTS:The protein expression ofα-SMA in model group was significantly higher than that in normal control group and blueberry prevention group(P<0.01).The protein expression of KLF11 in model group was significantly lower than that in normal control group and blueberry prevention group(P<0.01).The mRNA expression of KLF11 in blueberry prevention group was significantly higher than that in model group(P<0.01),while ColⅠmRNA in blueberry prevention group was significantly lower than that in model group(P<0.01).Compared with model group,the serum levels of ALT,AST,HA and PCⅢin other groups were significantly decreased(P<0.01),while the stage of liver fibrosis in blueberry prevention group was significantly lower than that in model group(P<0.01).CONCLUSION:Blueberry has certain preventive effect on hepatic fibrosis induced by carbon tetrachloride in rats,and its mechanism may be related to its effect on KLF11 expression.
