大豆肽减轻β-淀粉样蛋白肽诱导的小鼠原代海马神经元损伤

摘　　要：目的:研究大豆肽对β-淀粉样蛋白肽(Aβ)诱导的海马神经元损伤是否具有保护作用。方法:分离胚胎18.5 d C57BL/6小鼠海马原代神经元,培养7 d后用10μmol/L的Aβ25-35处理,制作神经元损伤模型;随后分别在神经元培养基中加入0.1%、0.5%和1.0%的大豆肽,处理48 h后,利用MTT法检测大豆肽对海马神经细胞活力的影响,DAPI染色检测神经细胞凋亡形态变化;免疫印迹检测神经细胞凋亡蛋白表达的变化;免疫荧光染色法检测原代神经元中微管蛋白β-Ⅲ-tubulin及微管相关蛋白MAP2的变化。结果:大豆肽提高了Aβ25-35所致神经损伤模型中海马神经元的细胞活力,大豆肽降低了Aβ25-35所致神经损伤模型中海马神经元的凋亡,减轻了细胞骨架的损伤。结论:大豆肽对Aβ诱导的神经细胞凋亡及细胞骨架损伤具有明显的抑制作用。Objective:To investigate the neuroprotective effects of soybean peptide on Aβ-induced toxicity in mouse primary hippocampal neurons.Methods:Primary hippocampal neurons were isolated from C57BL/6 mouse at embryonic day 18.5,cultured for 7 d,and then treated with 10μmol/L Aβ25-35 to establish neuronal injury model.Subsequently,0.1%,0.5%,1%concentration of soybean peptide(SBP)was added to the neuron medium for 48 h.The effect of SBP on the viability of hippocampal neurons was detected by MTT assay.The changes of neuronal apoptosis were detected by DAPI staining.The apoptosis-related proteins were determined by Western blotting,and the expression of microtubule protein beta-Ⅲ-tubulin and MAP-2 was used as an index for evaluating the morphological changes of neuronal cytoskeleton by using immunofluorescence staining.Results:SBP promoted the activity of hippocampal neurons with Aβ-induced injury,reduced the cell apoptosis,and repaired the cytoskeleton damage.Conclusion:SBP has obvious repairing and protective function against apoptosis and cytoskeleton damage induced by the neurotoxicity of Aβ.
