摘 要:红花油中含有大量的亚油酸,是制备亚油酸乙酯的一种良好原料。通过考察固定化脂肪酶催化红花油乙醇醇解反应从而优化酶的催化条件。实验结果表明:实验室自制固定化脂肪酶有较好的催化活性。通过单因素实验和正交试验,考察了反应溶剂、醇油添加比和温度等因素对固定化脂肪酶催化红花油乙醇醇解反应的影响,确定了酶催化反应最优工艺条件,在石油醚溶剂反应体系中(红花油和乙醇体积比4∶1)反应18 h,醇解转化率达到98.8%。经气质联用分析,酶催化产物中亚油酸乙酯相对质量分数为68.5%。Safflower oil contains a large amount of linoleic acid, which is a good raw material for preparing ethyl linoleate. In this study, alcoholysis of safflower oil catalyzed by immobilized lipase was studied to optimize the reaction conditions. The results indicate that the immobilized lipase shows good catalytic activity to the alcoholysis of safflower oil. The effects of reaction parameters including solvents, addition ratio of alcohol to oil and temperature were systematically investigated through single factor and orthogonal experiments, based on which the optimum parameters for the alcoholysis reaction were decided. Under the optimum conditions(with petroleum ether as solvent and the volume ratio of alcohol to safflower oil is 4∶1), the alcoholysis conversion rate could reach 98.8% after 18 h. The gas chromatography-mass spectrometry analysis revealed that the relative content of ethyl linoleate in the reaction products was 68.5%.
