口腔疣状癌中PTEN的表达及其抑癌作用的初步研究

摘　　要：目的:探讨磷酸酶及张力蛋白同系物(PTEN)在口腔疣状癌(OVC)中的表达及其可能的抑癌机制。方法:收集20例口腔疣状癌、20例口腔鳞状细胞癌组织石蜡标本,以10例外伤患者的正常口腔黏膜组织作为对照;免疫组织化学染色技术(SABC法)检测其中PTEN和pAKT的表达,并分析两者相关性。结果:PTEN在正常黏膜、口腔疣状癌、口腔鳞状细胞癌组织中的表达依次下降(P<0.05)并伴随pAKT的表达依次升高(P<0.05);PTEN与pAKT的表达呈负相关。结论:PTEN可能通过抑制pAKT的表达影响口腔疣状癌的发生发展。Objective:To investigate phosphate and tensin homolog(PTEN)expression in oral verrucous carcinoma(OVC)and its probable role of tumor suppression.Methods:The immunoreactivity levels of PTEN,pAKT were evaluated by SABC method in 20 tissue samples of OVC,20 tissue samples of oral squamous cell carcinoma(OSCC)and 10 tissue samples of normal oral mucosa(NM,the control group).The connection between PTEN and pAKT was analysed by Spearman correlation.Results:The expression of PTEN in OVC group was lower than that of the NM group(P<0.05),but higher than that of OSCC group.The expression of pAKT in OVC group was higher than that of the normal group(P<0.05),but lower than that of OSCC group.The PTEN and pAKT expression in OVC group showed negative correlation(r=-0.454,P<0.05).Conclusion:PTEN may play an important role in the development of OVC by feedback control of pAKT.
