摘　　要：目的分析PAX8抗体在淋巴瘤中的表达情况,探讨PAX8抗体在淋巴瘤中表达的原因。方法采用免疫组化EnVision法检测PAX8鼠单克隆抗体(克隆号OTI6H8)PAX8兔单克隆抗体(克隆号EP298)及PAX5在30例B细胞淋巴瘤和15例T细胞淋巴瘤中的表达情况。结果在非肿瘤B细胞以及除浆细胞瘤外的成熟B细胞肿瘤的不同亚型和RS细胞中,PAX8鼠单克隆抗体和PAX5显示几乎完全一致的反应模式,而PAX8兔单克隆抗体则为阴性。在非肿瘤T细胞和T细胞淋巴瘤中,PAX8鼠单克隆抗体、PAX8兔单克隆抗体和PAX5均无免疫反应。结论在非肿瘤B细胞和B细胞淋巴瘤中PAX8阳性,可能是因为某些克隆号的PAX8抗体与PAX5的N-末端区域发生交叉反应,而不是真正的阳性。对于PAX8阳性、上皮标记物阴性且形态不典型的肿瘤,鉴别诊断应考虑B淋巴细胞增生性疾病。当使用同一PAX家族蛋白的抗体时,评估其免疫组化染色结果必须明确所用抗体的克隆号。Objective To study the expression of different PAX8 antibodies in lymphomas,and to explore the reasons for the expression.Methods Immunohistochemical EnVision method was used to detect the expression of PAX8 mouse monoclonal antibody(OTI6 H8),PAX8 rabbit monoclonal antibody(EP298)and PAX5 in 30 cases of B-cell lymphoma and 15 cases of T-cell lymphoma.Results We observed an identical pattern of immunostaining between PAX8 mouse monoclonal antibody(OTI6 H8)and PAX5 in non-neoplastic B cells as well as in various subtypes of mature B-cell neoplasms except plasmacytomas,and RS cells,while PAX8 rabbit monoclonal antibody(EP298)was negative.All non-neoplastic T cells and T-cell lymphomas were negative using PAX8 mouse monoclonal antibody(OTI6 H8)and PAX8 rabbit monoclonal antibody(EP298)and PAX5.Conclusions PAX8 positivity being detected in non-neoplastic B cell and B-cell lymphomas is likely due to cross-reactivity between some PAX8 antibodies and N-terminal region of PAX5,but not true expression.When one encountered a PAX8-positive,epithelial marker-negative and atypical neoplasm,consideration of a B-cell lymphoproliferative disorder should be included in the differential diagnosis.When using antibodies directed against proteins of the same PAX family,one must take into consideration the antibody’s clone used in the evaluation of immunostaining results.
