摘  要:采用传统平板培养法与宏基因组测序法,对4℃贮藏保鲜条件下金针菇(Flammulina velutipes)表面细菌数量及种类进行分析。传统平板培养法共分离到3个门6个属的细菌,其中美洲爱文氏菌属和乳酸乳球菌属的细菌在不同贮藏时间均被分离到。宏基因组测序法共分析鉴定出13个门80个属的细菌,主要为厚壁菌门的乳酸乳球菌属和魏斯氏菌属,变形菌门的耶尔森氏菌属。研究表明:4℃贮藏条件下,金针菇表面细菌群落结构具有丰富的多样性,随着贮藏时间延长,菌落总数不断增加,而细菌种类数目下降,乳酸乳球菌是金针菇4℃贮藏保鲜期间的优势菌。Using the traditional culture plate separation and the metagenomics sequencing method,exterior bacterial microflora on Flammulina velutipes fruiting bodies stored at 4℃in vacuum sealed polypropylene bags was studied.The results showed that a total of 6 genera belonging to 3 phyla were isolated from different storage time sampling by the traditional culture plate separation method;and the dominant taxa,Lactococcus and Ewingella,were identified throughout the duration of the entire tested storage time.By metagenomics sequencing,a total of 80 genera belonging to 13 phyla were identified,and the dominant taxa were Lactococcus and Weissella of Firmicutes,and Yersinia of Proteobacteria.During the storage at 4℃,a rich bacteria diversity was initially detected,and then the diversity decreased over time in contrast to an increasing overall bacterial population.During the entire preservation period,the dominant and the major spoilage bacterium was Lactococcus lactis.
