摘 要:目的探讨KIF11、RhoA和Fascin蛋白在口腔鳞状细胞癌中的表达及意义。方法选取口腔鳞状细胞癌组织标本70例,选取癌旁组织(距癌组织>2 cm)作为对照,采用免疫组织化学染色法检测KIF11、RhoA和Fascin蛋白的表达;采用Real-time PCR检测KIF11、RhoA和Fascin mRNA的表达;比较其与临床病理特征的关系。结果口腔鳞癌组织KIF11、RhoA和Fascin蛋白阳性表达率分别为68.57%、60.00%和64.29%,均显著高于癌旁组织(P<0.05);鳞癌组织中KIF11、RhoA和Fascin mRNA的相对表达量分别为0.125±0.067,0.108±0.052和0.116±0.061。与癌旁组织相比,鳞癌组织中KIF11、RhoA和Fascin mRNA的表达量显著提高(P<0.05);KIF11、RhoA和Fascin蛋白与中低分化、有淋巴结转移、临床分期显著相关。KIF11、Fascin与RhoA蛋白表达均呈正相关(P<0.05)。结论KIF11、RhoA和Fascin蛋白在口腔鳞癌中呈高表达,与肿瘤分化程度、肿瘤直径等临床病理特征有关,提示KIF11、RhoA和Fascin蛋白可能参与了口腔鳞癌的发生与发展,联合检测可为临床上口腔鳞癌的诊断及预后提供了新的思路。Objective To investigate the expression and significance of KIF11,RhoA and Fascin proteins in oral squamous cell carcinoma.Methods 70 oral squamous cell carcinoma specimens were selected.The expression of KIF11,RhoA and Fascin protein was detected by immunohistochemical staining.The expression of KIF11,RhoA and Fascin mRNA was detected by real-time PCR.The relationship between the expression of KIF11,RhoA and Fascin and the clinicopathological characteristics was compared.The data were analyzed by SPSS 19.0 software.Results The positive expression rates of KIF11,RhoA and Fascin protein in oral squamous cell carcinoma were 68.57%,60.00%and 64.29%respectively,which were significantly higher than those in adjacent tissues(P<0.05);the relative expression levels of KIF11,RhoA and Fascin mRNA in oral squamous cell carcinoma were 0.125±0.067,0.108±0.052 and 0.116±0.061,respectively.Compared with the adjacent tissues,the expression of KIF11,RhoA and Fascin mRNA in squamous cell carcinoma was significantly increased(P<0.05),and KIF11,RhoA and Fascin protein were significantly correlated with middle and low differentiation,lymph node metastasis and clinical stage.KIF11,Fascin and RhoA protein expression were positively correlated(P<0.05).Conclusion KIF11,RhoA and Fascin proteins are highly expressed in oral squamous cell carcinoma,which is related to the degree of differentiation and the diameter of tumors,suggesting that KIF11,RhoA and Fascin proteins may be involved in the occurrence and development of oral squamous cell carcinoma.The combined detection of KIF11,RhoA and Fascin proteins may provide new ideas for the diagnosis and prognosis of oral squamous cell carcinoma.
