摘 要:为了对肥肝用骡鸭肝脏组织RNA-Seq数据特征进行分析,试验利用Trinity等软件和Samtools等工具对肝脏组织转录组数据进行基因表达鉴定,并对表达的基因进行单核苷酸多态性(single nucleotide polymorphisms,SNPs)、插入缺失标记(insertion-deletion,InDels)挖掘及功能分析。结果表明:在表达基因内可筛选出197 965个SNPs位点和93 990个InDels位点,这些基因涉及分子功能、生物学过程以及细胞组分三大类别的广泛生物学功能,SNPs所在基因除了富集在脂肪、能量代谢相关通路上,更多富集在免疫和内分泌等相关通路上。说明利用骡鸭肝脏组织转录组数据进行特征分析,筛选并探究表达基因的SNPs和InDels特征及相关基因的功能,可用于鸭肥肝生产相关基因的多态性检测、鸭肥肝形成机理等研究。The purpose of this study was to analyze the characteristics of the transcriptome data of foie gras tissue from mule ducks.Software such as Trinity and tools such as Samtools were used to identify gene expression in transcriptome data of liver tissue,and SNPs,InDels mining and functional analysis of related genes were performed for the expressed genes.Among the expressed genes,197965 SNPs loci and 93990 InDels loci could be screened out.Functional annotation and pathway analysis of these genes were involved in molecular function,biological process and broad biological functions of cell components in three categories.In addition to the accumulation of SNPs genes in fat and energy metabolism related pathways,more of them were mainly concentrated in immune and endocrine related pathways.In this study,the transcriptome data of theliver tissues from mule ducks were used for characteristic analysis,screening and exploring the characteristics of SNPs and InDels expression genes and the functions of related genes.It can be used for the polymorphism detection of related genes and the formation mechanism of foie gras in ducks.
