Objective To study the protective effect of Ginkgo biloba extract 50( GBE50) on vascular endothelial cells in a high glucose environment. Methods MTT assays were utilized to examine the HUVECs cell viability after the treatment of GBE50,changes in glutathione content were assessed by using the 7-amino-4-chloromethylcoumarin( CMAC)fluorescent stain technique, the effects of reactive oxygen species and GBE50 on autophagy were investigated by flow cytometry. Results MTT assay result showed that GBE50 significantly improved the viability of HUVECs in a high glucose environment. Flow cytometry showed that ROS-positive HUVECs were increased significantly in the high glucose environment, but GBE50 significantly reduced ROS production. GBE50 significantly increased the GSH content in HUVECs in the high glucose environment and decreased the ratio of MDA-positive cells. Immunofluorescence and western blot result showed that GBE50 significantly decreased the level of autophagy in HUVECs. Conclusions GBE50 reduces the effects of a high glucose environment on the activity of vascular endothelial cells by reducing autophagy and ROS production.
