摘  要:注射用中长链结构甘油三酯制备过程中,以大豆油和中链油为原料,采用脂肪酶TLIM催化合成中长链结构酯。在单因素试验优化的基础上,以酶用量、反应温度和时间为影响因子,羟值为响应值,采用响应面法优化合成工艺。结果表明,最佳工艺条件为酶用量2%、反应时间4 h、反应温度62℃,羟值预测值10.45,羟值实测值10.77,中长链结构酯中的甘油二酯含量为4.80%。本研究建立响应面优化模型,发现其拟合效果较好,可有效降低对中长链结构酯酶法合成中的副产物生成量。Soybean oil and medium chain oil as raw materials, are used by lipase TL IM enzymatic synthesis for the preparation of medium and long chain structural triglycerides of injection. Based on the optimization of the single factor experiment, the synthesis reaction process method was optimized by response surface methodology that was employed by the amount of enzyme, reaction temperature and time as the influencing factors, the hydroxyl value as the response value. The results showed that the optimal process conditions were: enzyme usage of 2 %;reaction time of 4 h;reaction temperature of 62 ℃;a predicted hydroxyl value of 10.45;a measured hydroxyl value of 10.77;The diglyceride content of the medium and long chain esters was 4.80 %. In this study, a response surface optimization model for enzymatic synthesis process was fit well with the measured value, which could be effectively reduced the content of by-products of medium and long-chain structural triglycerides in enzymatic synthesis.
