摘 要:以采自河北承德地区的一种野生白块菌为实验材料,运用形态学观察和分子生物学方法进行物种鉴定,确定河北白块菌的分类地位;制备河北白块菌水层、乙醇层和石油醚层提取物,对其进行总抗氧化能力、DPPH自由基清除能力和羟自由基清除能力的测定和研究,并与云南黑块菌进行比较分析。结果表明:河北白块菌为马蒂奥洛块菌属地菇状马蒂奥洛块菌(Mattirolomyces terfezioides)。河北白块菌提取物抗氧化活性均高于云南黑块菌。其中水层提取物抗氧化活性最强,每毫升含1g河北白块菌的水层提取物总抗氧化能力相当于14.4225mg/mL的VC标准品,是黑块菌水层提取物总抗氧化能力的2.29倍;每毫升含0.1g河北白块菌的水层提取物DPPH自由基清除能力为51.90%,显著高于云南黑块菌水层提取物;每毫升含0.01g河北白块菌的水层提取物羟自由基清除能力为31.35%,是云南黑块菌的1.32倍,两种块菌水层提取物的羟自由基清除能力均强于100μg/mL VC标准品,河北白块菌是其1.49倍,云南黑块菌是其1.13倍。A truffle has been taken as experimental material which is from Chengde area in Hebei province,and it has been identified basing on the morphological and molecular methods to determine the taxonomic status.Ethanol immersion,ultrasonic assisted extraction and organic solvent extraction are used to prepare the water layer,ethanol layer and petroleum ether layer extracts of Hebei white truffles.And the total antioxidant capacity,DPPH radical-scavenging ability and hydroxyl radical-scavenging ability of these extracts have been studied simultaneously to comparative with Yunnan black truffles.The results show that white truffles from Hebei province belong to Mattirolomyces terfezioides.The antioxidant activities of Hebei white truffles extracts are higher than that of Yunnan black truffles.The antioxidant activities of extracts from the water layer are the highest.The total antioxidant capacity of the water layer extracts containing 1g of Hebei white truffles per milliliter is equivalent to 14.4225 mg/mL of the VC standard,which is 1.29 times higher than that of the black truffles.DPPH radical-scavenging ability of the water layer extracts containing 0.1g of Hebei white truffles per milliliter is 51.90%,which is significantly higher than that of the Yunnan black truffles.Hydroxyl radical-scavenging ability of the water layer extracts containing 0.01g of Hebei white truffles per milliliter is 31.35%,which is 0.32 times higher than that of Yunnan black truffle.The hydroxyl radical-scavenging ability of the two kinds of truffle water layer extract is higher than that of 100 μg/mLVC standard.Hebei white truffle is 0.49 times higher than of it,Yunnan black truffle is 0.13 times higher than of it.
