摘 要:为探究新疆圆柏总黄酮(Flavonoids from Sabina vulgalis Ant.,SVFDs)的抗氧化活性,以及在食品保鲜中的应用;将20 mg/ml SVFDs梯度稀释为100μg/mL、50μg/mL、25μg/mL、12.5μg/mL浓度,抗坏血酸Vc为阳性对照,同时设置空白对照,阴性对照.FRAP法测定SVFDs总抗氧化活性,铁氰化钾法测定总还原力;检测不同浓度SVFDs对ABTS自由基、DPPH自由基、羟基自由基的清除作用;以小番茄果(Lycopersivon esculentum Mill.)和草莓(Fragaria×ananassa Duch.)为实验材料,检测SVFDs对超氧化物歧化酶SOD活性、丙二醛MDA含量的影响.结果显示,SVFDs具有显著的抗氧化活性,且随着浓度升高,抗氧化活性能力增强;虽然低浓度SVFDs还原能力不显著,但随着SVFDs浓度的升高,还原能力逐渐增强;同时SVFDs能够不同程度的清除ABTS自由基、DPPH自由基、羟基自由基活性;半抑制浓度IC50值分别是188.4μg/mL、2.251μg/mL、1 428.57μg/mL.保鲜实验结果显示,SVFDs可提高小番茄果、草莓中SOD的活性,显著降低MDA的含量.研究表明新疆圆柏总黄酮具有较强的抗氧化活性,选择性清除自由基能力,延长食品贮藏时间,作为果蔬保鲜剂,具有一定的开发潜力.In order to explore the antioxidant activity of flavonoids from Sabina vulgalis Ant.(SVFDs) in Xinjiang in vitro,and its application in food preservation,20 mg/ml SVFDs was diluted by gradient to 100 mg/mL,50 mg/mL,25 mg/mL and 12.5 mg/mL Ascorbic acid(Vc) was the positive control.Meanwhile,blank control and negative control were set.Total antioxidant activity of SVFDs was measured by FRAP method,total reduction force was measured by potassium ferricyanide method.The scavenging effect of SVFDs on ABTS free radical,DPPH free radical and hydroxyl free radical was tested,respectively.The effects of SVFDs on SOD activity and MDA content in Lycopersivon esculentum Mill and Fragariax ananassa Duch.were investigated.The results showed that SVFDs had significant antioxidant activity in a dose-dependent manner.Although the reduction ability of SVFDs at low concentration was not significant,with the increase of SVFDs concentration,the reduction ability was gradually enhanced.At the same time,SVFDs could scavenge ABTS free radical,DPPH free radical and hydroxyl free radical,with different scavenging effects.The IC50 values were 188.4μg/mL,2.251μg/mL and1 428.57μg/mL,respectively.The results showed that SVFDs could increase the activity of SOD and decrease the content of MDA in Lycopersivon esculentum Mill and Fragaria×ananassa Duch.It is concluded that the flavonoids from Sabina vulgalis Ant.(SVFDs) has strong antioxidant activity,selective free radical scavenging ability and prolonged the storage time of food As a fruit and vegetable fresh-keeping agent,SVFDs has a certain development potential.
