副溶血性弧菌标准物质制备的研究

摘　　要：为研究用冷冻干燥法制备含鱼肉基质的副溶血性弧菌标准物质,本试验以冷冻干燥过程中的保护率为指标,通过正交试验选择保护率最高的复方保护剂。结果表明:以钴-60辐照灭菌的鱼肉粉作为基质,按适当的比例与复方保护剂混合,添加预定数量的菌液,于-70℃速冻24 h后进行冷冻干燥,制备成的干粉状标准物质均匀性良好。冷冻干燥样品在-70℃条件下保存360 d,特性值稳定。This experiment studies the method of the Vibrio Parahemolyticus reference materials containing fish substrate by the freeze-drying preparation.For indicators of protection formula in the process of freeze-drying through orthogonal test to the choose freeze-dried protection scheme of highest protectant formula.The results showed that the fish powder sterilizated by Cobalt-60 irradiation as substrate,the appropriate proportion mixed with compound protective agent,adding reservation number of bacteria liquid,which was freeze-drying after-70℃quiok freezing for 24 h.The homogeneity of the dry powder reference materials was good.Freeze-dried samples preserved in the condition of-70℃for 360 days,the characteristic value was stable.
