摘　　要：目的:探究PTEN基因突变及mRNA表达与卵巢癌病理生物学特征的关系。方法:选取2017年5月-2018年5月上皮性卵巢囊肿患者10例、卵巢交界性肿瘤患者18例、卵巢癌患者120例及卵巢正常者10例。全部以PCR-SSCP法进行PTEN外显子5突变检测,比较四组卵巢组织的mRNA表达;分析PTEN基因突变、mRNA表达和卵巢癌病理生物学特征的关系。结果:上皮性卵巢癌患者发现有PTEN外显子5突变;卵巢交界性肿瘤、卵巢癌患者在PTEN基因的mRNA表达上低于上皮性卵巢囊肿与正常卵巢,差异有统计学意义(P<0.05)。PTEN基因的mRNA表达和卵巢癌病理分期之间为负相关联系,与组织分化程度为正相关联系。结论:PTEN外显子5突变及mRNA下调与卵巢癌的病理生物学行为变化有着较为密切的联系,PTEN基因可以用于反映卵巢癌的病理生物学特征,临床可以借此对卵巢癌进行准确诊断。Objective:To explore the relationship in PTEN gene mutation and mRNA expression and pathobiological characteristics of ovarian cancer.Methods:From May 2017 to May 2018,10 patients with epithelial ovarian cyst,18 patients with borderline ovarian tumors,120 patients with ovarian cancer and 10 patients with normal ovary were selected.The PTEN exon 5 mutation was detected by PCR-SSCP method to compare the mRNA expression of ovarian tissue in four groups.To analyze the relationship in PTEN gene mutation,mRNA expression and the pathobiological characteristics of ovarian cancer.Results:PTEN exon 5 mutation was found in patients with epithelial ovarian cancer.Patients with borderline ovarian tumors and ovarian cancer had lower mRNA expression of PTEN genes than epithelial ovarian cysts and normal ovarian cysts,the differences were statistically significant(P<0.05).There was a negative correlation between mRNA expression of PTEN gene and pathological stage of ovarian cancer and a positive correlation with the degree of tissue differentiation.Conclusion:PTEN exon 5 mutation is closely related to the down-regulation of mRNA and the pathobiological characteristics of ovarian cancer,PTEN genes can be used to reflect the pathological and biological characteristics of ovarian cancer,which can be used to accurately diagnose ovarian cancer.
