摘  要:采用分步醇沉法得到4种银耳多糖级分--STP 20、STP 40、STP 60、STP 80,分析其得率、纯度和相对分子质量及纯化STP 60的单糖组成,研究其对酪氨酸酶的抑制作用及作用类型。结果表明,STP 60的得率和纯度均为最高,分别为19.28%和81.12%;4种银耳多糖级分均具有一定酪氨酸酶抑制作用,其中STP 60的抑酶作用最大,质量浓度1 mg/mL时对酪氨酸酶活性的抑制率为37.55%;STP 60对酪氨酸酶的抑制作用属于非竞争性可逆抑制。银耳多糖在美白类化妆品中具有潜在应用价值。Four Tremella fuciformis polysaccharide fractions defined as STP 20,STP 40,STP 60 and STP 80 were obtained by stepwise alcohol precipitation.Their yields,purity and relative molecular weight and the monosaccharide composition of purified STP 60 were analyzed.Moreover,their inhibition on tyrosinase activity was investigated.The results showed that STP 60 had the highest yield(19.28%)and purity(81.12%)in the four fractions.All the polysaccharide fractions demonstrated certain inhibitory effect on tyrosinase activity,of which STP 60 exerted the highest inhibition,the activity of tyrosinase was inhibited by 37.55%at 1 mg/mL.STP 60 inhibited the enzyme by non-competitive reversible inhibition way.Therefore,Tremella fuciformis polysaccharide possess the potential to be applied in skin whitener cosmetics.
