摘 要:黄酮类化合物是一类具有广泛生物活性的植物次生代谢产物,具有潜在的应用前景。然而,因大多数黄酮存在水溶性差、透水性差而导致生物利用率低,限制了其应用。文章以常见的黄酮类化合物山奈酚为原料,制备了新型具有良好水溶性的山奈酚磺基衍生物(山奈酚-3’,8-二磺酸钠)及其铜(Ⅱ)配合物。采用核磁共振氢谱和电喷雾质谱确定了山奈酚-3’,8-二磺酸钠分子结构,用元素分析、红外光谱、紫外可见光谱以及热重分析等方法对配合物进行了表征,揭示配合物的配位比为1:1,且具有变形五配位结构。同时,着重研究了山奈酚-3’,8-二磺酸钠及其铜(Ⅱ)配合物对DPPH自由基、羟基自由基、超氧阴离子自由基和脂质过氧化的抑制作用。结果表明,主题铜(Ⅱ)配合物较山奈酚-铜配合物具有更优良的抗氧化活性。Flavonoids are a class of plant secondary metabolites with a wide range of biological activities,and have good prospects for application.However,application of the most flavonoids is limited by their low bioavailability resulting from poor water solubility and permeability.In this paper,a new and highly water-soluble sulfated derivative of kaempferol (kaempferol-3’,8-disulfonic acid sodium) and its copper (Ⅱ) complex were prepared from the common flavonoid kaempferol.The structure of the sulfonated derivative was determined by ~1H nuclear magnetic resonance and electrospray ionization mass spectrometry,and the complex was characterized by elemental analysis,infrared spectroscopy,UV-Vis absorption spectroscopy and thermogravimetric analysis.The results showed that the complex had a distorted five-coordinated geometry with the coordination ratio of 1:1.More importantly,the inhibition of kaempferol-3’,8-disulfonic acid sodium and its copper (II) complex for DPPH free radical,hydroxyl radical,superoxide anion radical and lipid peroxidation was studied.It was found that the copper complex has better antioxidant activity than kaempferol copper complex.
