摘  要:目的利用全基因组测序技术构建稀有细菌Leifsonia flava SYP-B2174~T的基因组草图,解析其基因特性,挖掘次级代谢产物基因的资源。方法以Illumina HiSeq PE150测序平台对菌株SYP-B2174~T进行全基因组测序,使用生物信息软件对测序数据进行基因组组装、基因预测与注释。使用琼脂扩散法对菌株的代谢产物进行抗菌活性测定。结果菌株SYP-B2174~T基因组测序得到16个contig,总长为3 782 833 bp,G+C含量为68.6 mol%,注释到3 543个基因,包含3 455个蛋白,46个tRNA,3个其他RNA,35个假定蛋白,GenBank登录号为SPQZ00000000。预测得到聚酮类、非核糖体多肽样合酶和萜类化合物的生物合成基因簇,与现有基因簇相似度≤21%。拮抗实验结果表明菌株SYP-B2174~T的代谢产物对6种人类致病菌的生长有抑制作用。结论 Leifsonia flava中具有众多次级代谢产物的合成基因簇且与已知基因簇相似度低,并且代谢物具有抗菌活性,可为今后开展活性化合物分类鉴定、基因改造、合成生物学的研究奠定基础。Objective Drafting of genome map by next-generation sequencing technology,to analysis the genetic information of rare bacteria Leifsonia flava SYP-B2174~T and explore the genetic resources of secondary metabolites.Methods The whole genome of SYP-B2174~T strain was sequenced on the IlluminaHiSeq PE150 platform,assembled by SOAPdenovo,genes predicted by GeneMarkS,and genes annotated by Blast.Antibacterial activities of secondary metabolites were tested against human pathogenic bacteria by employing the disc diffusion method.Results The strain SYP-B2174~T obtained 16 contigs,total length was 3 782 833 bp.The genomic G+C content was determined to be 68.6 mol%.It contained 3 543 genes,3 455 proteins,46 tRNAs,3 other RNAs and 35 pseudogenes.The Whole Genome Shotgun project had been deposited at DDBJ/ENA/GenBank under the accession SPQZ00000000.Gene clusters of PKS,NRPS-like fragment and terpene were annotated by antiSMASH,and the similarity with existing gene clusters was less than 21%.Results of antibacterial activities displayed that secondary metabolites of strain SYP-B2174~T had the activity against Pseudomonas aeruginosa,Escherichia coli,Klebsiella pneumonia,Salmonella paratyphi B,Staphylococcus aureus and Bacillus cereus.Conclusion There are many synthetic gene clusters of secondary metabolites in Leifsonia flava,which have low similarity with known gene clusters.And the metabolites have antimicrobial activity,which can guide future research on identification of active compounds,genetic modification and synthetic biology.
