摘 要:目的探究HPV52型E6和E7基因突变位点分布情况,分析宫颈癌以及癌前病变同基因突变之间的关系。方法选择某院2018年1月8日-2019年11月8日宫颈细胞检测的240例标本作为研究样本,对人乳头瘤病毒52型E6以及E7基因进行聚合酶联反应扩增。对扩增产物进行碱基序列以及蛋白序列检测,了解其基因突变位点分布情况。结果有208例经扩增后出现人乳头瘤病毒52型E6以及E7基因序列,并且E6基因存在16种突变位点,E7基因存在17种突变位点。不同突变位点在突变率方面比较差异无统计学意义(P>0.05)。Ⅰ+型样本中人乳头瘤病毒52型E6基因突变率明显低于正常样本(P<0.05);Ⅰ+型样本中人乳头瘤病毒52型E7基因突变率同样低于正常样本,但2者比较差异不存在统计学意义(P>0.05)。结论人乳头瘤病毒52型E6基因突变率低于正常样本,当E6基因发生突变时可能会降低人乳头瘤病毒致癌作用。Objective To explore the distribution of E6 and E7 gene mutations of HPV52,and analyze the relationship between cervical cancer,precancerous lesions and gene mutations.Methods 240 samples of cervical cells tested in hospitals were selected as research samples,and the test time was between January 8,2018 and November 8 th,2019.The E6 and E7 genes of human papillomavirus 52 were amplified by PCR.Base sequence and protein sequence of the amplified products were detected to understand the distribution of gene mutation sites.Results There were 208 cases of HPV 52 E6 and E7 gene sequence after amplification,and there are 16 mutation sites in E6 gene and 17 mutation sites in E7 gene.There was no significant difference in mutation rate among different mutation sites(P>0.05).The mutation rate of hpv52 E6 gene in typeⅠ+was significantly lower than that in normal samples(P<0.05);The mutation rate of hpv52 E6 gene in typeⅠ+samples was also lower than that in normal samples,but there was no significant difference between them(P>0.05).Conclusions The mutation rate of E6 gene of human papillomavirus 52 was lower than that of normal samples.When E6 gene mutation occurred,it might reduce the carcinogenesis of human papillomavirus.
