摘  要:运用基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS),对2种云南特色高值野生食用菌块菌和美味牛肝菌进行物种甄别鉴定。运用液氮甲酸法处理样品,获得浓度分别为0.243 mg·mL^-1和0.284 mg·mL^-1的块菌和美味牛肝菌的蛋白质提取液,经MALDI-TOF-MS采集指纹图谱,对块菌和美味牛肝菌的蛋白指纹图谱进行比对与聚类分析。结果表明,产地不同的同种野生食用菌具有相似的蛋白指纹图谱和物种特征质谱峰,而不同种类的食用菌间具有差异明显的特征蛋白指纹图谱。MALDI-TOF-MS可以用于云南出口高值特色野生食用菌块菌和美味牛肝菌的物种快速甄别。Two edible fungi of Tuber indicum Cooke&Massee and Boletus bainiugan Dentinger were classified and identified by matrix-assisted laser desorption ionization time-of-flight mass spectrometry MALDI-TOF-MS technology.The samples were treated with liquid nitrogen formic acid,and the protein extracts of T.indicum and B.bainiugan were obtained with concentrations of 0.243 mg·mL^-1 and 0.284 mg·mL^-1,respectively.The protein fingerprints of T.indicum and B.bainiugan were compared and cluster analyzed by MALDI-TOF-MS.The results showed that the same kind of wild edible fungi from different areas had similar protein fingerprints and species characteristic mass spectrum peaks,but different kinds of edible fungi had different characteristic protein fingerprints.MALDI-TOF-MS can be used to identify the high value species of T.indicum and B.bainiugan in Yunnan province.
