摘  要:建立pH示差法测定红花羊蹄甲花瓣中花色苷的含量。用酸性乙醇溶液从红花羊蹄甲花瓣中提取花色苷,确定提取液的最大吸收波长、缓冲液pH、反应平衡时间、反应平衡温度,在此条件下进行精密度试验和加标回收率试验。结果表明提取液的最大吸收波长为535 nm,缓冲液pH为1.0和4.5,反应平衡时间为50 min,反应平衡温度为40℃。测定结果的相对标准偏差为1.17%(n=6),花瓣中花色苷含量为10.401 mg/g,加标回收率为99.36%,RSD为1.69%。该方法准确可靠,可以用于红花羊蹄甲花瓣中花色苷含量的测定。It is to develop pH differential method for determining the contents of anthocyanins in petals of Bauhinia blakeana.Anthocyanins was extracted by acid ethanol from petals of Bauhinia blakeana.The maximum absorption wavelength of the extracting solution,the pH of the buffer solution,the time of the reaction equilibrium and the reaction equilibrium temperature were determined,and precision and recovery test were carried out under these conditions.The results showed that the maximum absorption wavelength of the extracts was 535 nm,the pH was selected as 1.0 and 4.5,the reaction equilibrium time was 50 min,and the reaction equilibrium temperature was 40℃.The contents of anthocyanins from petals of Bauhinia blakeana was 10.401 mg/g,the RSD was 1.17%,the recovery rate was 99.36%,and the RSD was 1.69%.This method is accurate and reliable,and can be used for quantitative analysis of anthocyanins from the petals of Bauhinia blakeana.
