摘 要:目的 12-脂氧合酶(12-lipoxygenase,12-LOX)是花生四烯酸代谢通路的关键酶,近年来其与肿瘤发生发展的相关性研究备受关注。本研究探讨12-LOX在结直肠癌组织中的表达及其临床意义,以及12-LOX抑制剂黄芩素(baicalein,BAI)对其表达的影响。方法选取2010-06-01-2017-06-01天津市公安医院结直肠癌手术治疗患者80例。收集癌组织(肠癌组)和配对正常组织标本距离边缘5cm(正常组),另选同期60例腺瘤组织标本(腺瘤组),分析12-LOX表达与结直肠癌患者临床病理特征相关性。应用免疫组织化学PV-6000法检测;实时荧光定量PCR(real-time quantitative polymerase chain reaction,RT-qPCR)技术检测黄芩素对HT-29细胞中12-LOX mRNA表达的影响。采用SPSS 19.0对数据进行统计学分析。结果 12-LOX表达率正常组为13.75%(11/80)、腺瘤组为28.33%(17/60)、肠癌组为85.00%(68/80),12-LOX表达在肠癌组与腺瘤组(χ2=46.158,P<0.05)、肠癌组与正常组(χ2=81.238,P<0.05)、腺瘤组与正常组(χ2=4.557,P<0.05)两两比较,差异有统计学意义。12-LOX表达与结直肠癌淋巴结转移(χ2=15.703,P<0.001),Dukes分期(χ2=7.816,P=0.009)有关联。HT-29细胞中12-LOX mRNA相对表达量干预36h20μmol/L黄芩素组为0.692 7±0.091 7,40μmol/L组为0.680 0±0.056 4,80μmol/L组为0.480 3±0.072 0,均低于空白对照组的1.081 1±0.149 1,差异有统计学意义,均P<0.05;干预24h80μmol/L黄芩素组为0.699 3±0.047 5,低于空白对照组的1.021 5±0.092 8,差异有统计学意义,P<0.05。双因素方差分析显示,主效应药物、时间以及药物×时间对HT-29细胞12-LOX mRNA的表达均有影响(F药物=8.450,F时间=29.461,F药物×时间=4.110,均P<0.05),其中时间的影响更明显。两两多重比较中,主效应时间的各作用时间对HT-29细胞12-LOX mRNA的表达影响差异均有统计学意义(均P<0.05),主效应药物的80μmol/L黄芩素组与其他剂量组比较,差异均有统计学意义,均P<0.05。结论 12-LOX在结直肠癌�OBJECTIVE 12-lipoxygenase(12-LOX)is one of the key enzymes in arachidonic acid metabolism pathway.In recent years,the relationship between 12-LOX and tumorigenesis has attracted much attention.This study aimed to investigate the expression of 12-LOX in colorectal cancer(CRC)and analyze its potential clinical significance,and to observe the effect of baicalein(BAI)on the expression of 12-LOX mRNA in HT-29 cell.METHODS The expression levels of 12-LOX in 80 CRC tissues along with the cognate 80 paired adjacent normal tissues,that received surgical treatment from 2010-06-01 to 2017-06-01 in the Public Security Hospital of Tianjin and 60 cases of colorectal adenoma tissues from the same period were analyzed by immunohistochemistry(PV-6000 method).The correlations between the expression of12-LOX and the clinical pathological characteristics of CRC were also analyzed.RT-qPCR methods were used to examine the expression of 12-LOX mRNA by the effect of BAI in HT-29 cell.SPSS 19.0 statistical softwave was used to analyze the date.RESULTS The expression rates of 12-LOX were 13.75%(11/80)in normal group,28.33%(17/60)in adenoma group and 85.00%(68/80)in CRC group.There were difference in the expression rates of 12-LOX between CRC group and adenoma group(χ2=46.158,P<0.05),between CRC group and normal group(χ2=81.238,P<0.05),between adenoma group and normal group(χ2=4.557,P<0.05).The expression of 12-LOX were correlated with lymph node metastasis(χ2=15.703,P<0.001)and Dukes stage(χ2=7.816,P=0.009).The relative folds of 12-LOX mRNA in HT-29 cells were 0.6927±0.0917 in 20μmol/L baicalein group for 36 hours,0.6800±0.0564 in 40μmol/L baicalein group for 36 hours,0.4803±0.0720 in 80μmol/L baicalein group for 36 hours,which were lower than 0μmol/L baicalein group for 36 hours(1.0811±0.1491),all P<0.05.The relative fold of 12-LOX mRNA in HT-29 cells was 0.6993±0.0475 in 80μmol/L baicalein group for 24 hours,lower than 0μmol/L baicalein group for 24 hours(1.0215±0.0928),P<0.05.Two-way analysis of variance showed that th
