摘  要:本文采用流式细胞仪,研究了扩培酵母细胞的周期变化规律,确定多级扩培可使酵母的细胞生长周期趋向同步,并确定啤酒酵母的细胞周期为6h左右。同时研究了扩培酵母不同转接时间点对酵母生长情况的影响,确定选取酵母处于对数生长期以G1期占优的时间点进行扩培转接,既能保证下一步扩培有足够基数的酵母确保扩培速度,又可确保酵母在转接过程中受到的损伤降到最低,逐级扩培的转接时间在18-24h内最佳。该研究为啤酒厂实际生产中酵母扩培转接时间提供理论依据,具有重要意义。In this paper,flow cytometry was used to study the changing regularity of cell cycle of beer ycast in propagation.It was determined that the tendency of the cell cycle of beer ycast would be synchronized by multi-stage propagation,and the cell cycle of beer yeast was about 6h.At the same time,the effects of different transfer time points of expanded yeast on the growth of yeast were studied in this article.It was determined that the time points with more yeast cells in Gl stage of non-proliferation-period yeast in exponential growth were selected for next yeast expansion,which could not only ensure the yeast with enough number in the next expansion to ensure the expansion speed,but also ensure that the damage of yeast in the transfer process was minimized.The best transfer time was 18-24 hours in step by step propagation.This study provides a thcoretical basis for the transfer time of ycast culture expansion and it is a great significance in industrial breweries.
