摘 要:目的探讨RhoC-shRNA转染对口腔鳞癌细胞增殖及侵袭能力的影响。方法选取人口腔鳞癌细胞CAL-27细胞系进行培养,RhoC-shRNA-LVRU6MP慢病毒表达载体转染CAL-27,同时转染shRNA对照序列的空载体LVRU6MP作为对照组,嘌呤霉素筛选稳定表达RhoC-shRNA的细胞系。通过RT-PCR检测敲低效果。分别通过MTT实验、细胞克隆形成实验、划痕实验、Transwell实验观察RhoC基因敲低对人口腔鳞癌细胞CAL-27增殖,克隆和迁移,侵袭的生物学行为的影响。结果 RT-PCR结果显示与对照组相比,实验组RhoC的表达降低80%左右,MTT实验RhoC-shRNA组和对照组相比增殖能力明显降低(P<0.01),细胞克隆形成实验RhoCshRNA组和对照组相比细胞克隆能力明显降低(P<0.01),划痕实验RhoC-shRNA组和对照组相比细胞迁移能力明显降低(P<0.01),Transwell实验RhoC-shRNA组和对照组相比细胞侵袭能力明显降低(P<0.01)。结论抑制RhoC的表达可以抑制人口腔鳞癌细胞CAL-27的增殖、迁移和侵袭等恶性生物学行为能力。Objective To investigate the roles of RhoC gene in the human oral squamous cell carcinoma cell line CAL-27.Methods Human squamous cell carcinoma cell line CAL-27 was cultured in DMEM medium,and the RhoCshRNA-LVRU6MP lenti viral expression vector was transfected into CAL-27.Meanwhile,the control vector LVRU6MP of the shRNA control sequence was transfected into CAL-27 cells as the control group,and the cell line expressing RhoCshRNA was screened by puromycin.The knock down effect was detected by RT-PCR.Proliferation,cloning,migration and invasion of oral squamous cell carcinoma cells were observed by MTT assay,cell cloning formation assay,scratch assay and Transwell assay,respectively.Results RT-PCR results showed that compared with the control group,the expression of RhoC in the experimental group was reduced by about 80%.The proliferation ability of the RhoC-shRNA group in the MTT experiment was significantly decreased compared with the control group(P<0.01).The cell cloning ability of the RhoCshRNA group was significantly decreased compared with the control group(P<0.01).The cell migration ability of the RhoCshRNA group was significantly decreased compared with the control group(P<0.01).Transwell experiment RhoC-shRNA group significantly decreased cell invasion ability(P<0.01).Conclusion Inhibition of RhoC expression can inhibit the proliferation,migration and invasion of CAL-27.
