摘  要:以红曲为研究对象,研究红曲霉菌液体发酵液和固体红曲样品中桔青霉素的含量,采用HPLC检测方法对桔青霉素进行快速定量测定.并对桔青霉素的提取方法和测定的色谱条件进行了优化,进而验证了HPLC的精密度、稳定性、重复性、检出限和回收率.结果表明:HPLC检测条件为色谱柱SunFire C18(250×4.6/5μm)、流动相乙腈和水的体积比为67∶33、等度洗脱、柱温28℃、流速1.0 mL·min^-1、检测时间15 min、荧光检测器,激发波长为331 nm,发射波长为500 nm.桔青霉素的线性范围在0.050~10.000μg·mL^-1表现良好,R2=0.998 5,HPLC检测的精密度、稳定性和重复性良好,最低检出限1 ng·mL^-1,平均加标回收率99.35%,RSD值0.58%,检测时间更短,甲醇提取法与传统的红曲类产品提取方法相比提出率更高.Absrtact:In this study,the content of citrinin in Monascus liquid fermentation broth and solid Monascus samples was studied,and the rapid quantitative determination of citrinin was carried out by HPLC.The extraction method and chromatographic conditions of citrinin were optimized,and the precision,stability,repeatability,detection limit and recovery of HPLC were verified.The results showed that the detection conditions of HPLC were SunFire C18 column(250×4.6/5 μm),the mobile phase was an isocratic mobile phase of 67% acetonitrile,the column temperature was 28 ℃,the flow rate was 1.0 mL·min^-1,the detection time was 15 min,the fluorescence detector:excitation wavelength was set at 331 nm,emission wavelength was set at 500 nm.The linear range of citrinin was from 0.050 to 10.000 μg·mL^-1,R^2=0.998 5.The precision,stability and repeatability of HPLC were good.The minimum detection limit was 1 ng·mL^-1.The average recovery of citrinin was 99.35%,RSD value was 0.58%.The detection time is shorter,and the methanol extraction method has higher extraction rate than the traditional method of Monascus products.
