摘  要:目的利用公共平台的子宫内膜癌芯片数据筛选子宫内膜癌相关基因。方法下载GEO数据库的子宫内膜癌数据集GSE17025和GSE39099,筛选差异表达基因,利用DAVID软件对差异表达基因进行GO和KEGG信号通路富集分析。利用STRING在线工具对差异表达编码蛋白进行蛋白-蛋白相互作用网络分析,并采用Cytoscape进行可视化以及核心模块和关键基因的挖掘。结果初步筛选出子宫内膜癌相关上调基因101个,下调基因128个。上调基因主要位于胞核,富集于细胞增殖、蛋白质结合、细胞周期信号通路,下调基因主要位于胞外区域,富集于药物反应、金属离子结合、cAMP信号通路。成功构建差异表达基因编码蛋白的相互作用网络,利用MCODE算法筛选了两个核心模块,其中关键基因为DEPDC1和CXCL8。通过TCGA数据库的数据证实,两个关键基因均在子宫内膜癌中表达升高(P<0.05),且对患者的生存率有显著影响(P<0.05)。结论利用公共数据平台的子宫内膜癌多组芯片数据,筛选出两个关键基因DEPDC1和CXCL8,可能在子宫内膜癌中发挥着重要作用,为子宫内膜癌新的诊断标志物和治疗靶点筛选提供新思路。Objective To identify endometrial cancer related genes using the endometrial cancer microarray data from Gene Expression Omnibus(GEO)database.Methods The data sets GSE17025 and GSE39099 were downloaded to screen the significant genes differentially expressed in endometrial cancer.DAVID was used to perform the GO and KEGG signaling pathways enrichment of the common differentially expressed genes(DEGs).STRING was used to analyze the protein-protein interaction(PPI)network of DEGs,and Cytoscape was used for visualization and the mining of core modules and key genes.Results There were 101 up-regulated genes and 128 down-regulated genes selected for endometrial carcinoma.Up-regulated genes were mainly located in the cell nucleus and enriched in cell proliferation,protein binding,and cell cycle signaling pathways,while down-regulated genes were mainly located in extracellular regions and enriched in drug response,metal ion binding,and cAMP signaling pathways.The PPI network of differentially expressed gene was successfully constructed,and two core modules were screened by MCODE algorithm,among which the key genes were DEPDC1 and CXCL8.Data from TCGA database confirmed that both key genes were differentially expressed between endometrial cancer and normal tissue(P<0.05)and had a significant impact on the survival rate of patients(P<0.05).Conclusion Using multiple endometrial cancer microarray data from public database,DEPDC1 and CXCL8,two key genes,were screened out and were suggested to play an important role in endometrial carcinoma.The study provided an insight for novel diagnostic markers and therapeutic targets screening for endometrial cancer.
