摘  要:GLI作为Hedgehog(Hh)信号通路的关键调控因子,在各种癌症的发生发展中参与了重要的角色。本文总结了GLI在多种癌症发生中的作用及其促进肿瘤进展相关的机制,同时还讨论了GLI对肿瘤干细胞的调控功能。因此,靶向GLI抑制剂治疗可能给癌症患者带来了一种新思路。鉴于此,我们又总结了多种GLI抑制剂的作用特点以及分析了GLI抑制剂在体内外实验中的抗肿瘤效应。虽然目前只有3种GLI抑制剂进入了临床试验阶段,但未来会开展更多的临床试验来验证靶向GLI药物的临床疗效。GLI,as a key regulator of Hedgehog(Hh)signaling pathway,plays an important role in the carcinogenesis and progression of various cancers.This review summarizes the role of GLI in the tumorigenesis of a variety of malignancies and the mechanisms involved in promoting tumor progression.Meanwhile,it also discuss the regulatory functions of GLI on cancer stem cells.Therefore,targeting GLI inhibitors as a clinical therapy may bring novel insights to cancer patients.In view of these above points,it also summarizes the characteristics of several GLI inhibitors and their anti-tumor effects in vivo and in vitro.At present,there are only three GLI inhibitors participated in the clinical trials from different types of cancer,more clinical trials should be performed in the future urgently to verify the clinical efficacy of targeting GLI therapy.
