摘  要:目的探究菊粉(inulin)对化疗药物环磷酰胺(cyclophosphamide,CY)造成的肠黏膜免疫系统损伤小鼠的保护作用。方法 50只昆明种小鼠随机均分为5组:正常组,模型组(CY 50 mg/kg),低(200 mg/kg)、中(400 mg/kg)、高(600mg/kg)剂量的菊粉组。菊粉组持续灌胃菊粉28d,同模型组于第27d起连续2d灌胃相同剂量的环磷酰胺。观察小鼠的体重变化,检测胸腺指数、脾指数和派氏结(Peyer’spatches)值,对空肠和结肠段进行HE染色观察,并用免疫组织化学法观察空肠和结肠黏膜免疫球蛋白A阳性(IgA+)细胞数;ELISA法检测空肠、结肠段分泌型免疫球蛋白A(SIgA)的含量。结果与模型组相比,高剂量组小鼠第28 d体质量增加(P<0.05),中、高剂量组的免疫器官指数均明显增加(P<0.01);高剂量组的派氏结值明显高于模型组(P<0.01);模型组小鼠出现空肠绒毛萎缩、脱落伴炎细胞浸润等病变,绒毛长度与隐窝深度的比值、IgA+细胞数和SIgA分泌水平相较于正常组均明显下降(P<0.01)。与模型组相比,三个剂量组肠绒毛损伤均有改善,其中高剂量组绒毛结构接近正常组;高剂量组空肠和结肠的IgA^+细胞和SIgA分泌水平均明显高于模型组(P<0.01)。结论高剂量菊粉可以起到保护因环磷酰胺造成的小鼠肠黏膜免疫系统损伤的作用。[营养学报,2020,42(3):260-265]Objective To explore the potential protective effect of inulin on intestinal mucosal immunity in mice treated with cyclophosphamide(CY). Methods Fifty healthy KM mice were randomized into five groups: normal group, model group(CY 50 mg/kg), low-dose inulin group(200 mg/kg), middle-dose inulin group(400 mg/kg), high-dose inulin group(600 mg/kg). Normal saline or inulin was given by gavage once a day for 28 days. All groups except normal group were given CY(50 mg/kg/bwt) by gavage in the 27 th days. The body weight, immune organ index and Peyer’s patches value were measured. The micromorphology structure and the immunoglobulin A positive(IgA+) cells in jejunum and colon were observed respectively after hematoxylin-eosin(HE) staining and immunohistochemical staining. The contents of secretory immunoglobulin A(SIgA) in jejunum and colon mucus were detected by ELISA. Results At days 28, the body weight of mice in the high dose group was higher compared with the model group(P<0.05), and the immune organ indexes in the middle and high-dose groups were remarkably increased(P<0.01);the Peyer’s patches value in the high-dose group was significantly higher than that in the model group(P<0.01);the mice in the model group had atrophy and exfoliation in jejunal villus with inflammatory cell infiltration and other pathological changes;villus height to crypt depth ratio in jejunum, the number of IgA+ cells and the level of SIgA were significantly lower compared with the normal group(P<0.01). Compared with the model group, the jejunal villus injury were improved in the three inulin dose groups, in which the villus structure of the high dose group was close to that of the normal group;the IgA^+ cells and SIgA secretion levels in jejunum and colon of the high-dose group were significantly higher than those of the model group(P<0.01).
