鲜冬虫夏草抗氧化和醛糖还原酶抑制活性的研究

摘　　要：为评价鲜冬虫夏草的生理活性,对鲜冬虫夏草分别用水和95%乙醇进行加热回流提取,通过系统溶剂萃取和大孔吸附树脂HPD-100柱色谱法进一步获得不同的组分。对分离所得的各组分进行体外抗氧化和醛醣还原酶(AR)抑制活性研究。结果表明:鲜冬虫夏草水提物的95%乙醇洗脱物具有显著的抗氧化活性和抑制醛糖还原酶活性,水提物、粗多糖、乙酸乙酯萃取物、正丁醇萃取物和水层也表现出一定程度的抗氧化活性和醛糖还原酶抑制活性。这一研究说明鲜冬虫夏草可用于临床糖尿病并发症的预防和辅助治疗。In order to evaluate the physiological activity of fresh Ophiocordyceps sinensis, the isolation and in vitro activity tests have been performed. The fresh O.sinensis was extracted with hot reflux water and 95% ethanol to produce aqueous extract(Ⅰ) and alcohol extract(Ⅳ), respectively. The extract I has been isolated by the HPD-100 macroporous resin column chromatography to yield the aqueous fraction which was further purified by the removal of protein(Ⅱ) and ethanol fraction(Ⅲ), and the extract Ⅳ has been successively partitioned with petroleum ether, ethyl acetate and n-butanol in water to give four main fractions(Ⅴ—Ⅷ). Those fractions were tested in vitro their antioxidant and aldose reductase(AR) inhibitory activities. As results, the fraction Ⅲ showed the most potent antioxidant and aldose reductase inhibitory activities. Additionally, fractions Ⅰ, Ⅱ and Ⅵ—Ⅷ, also exhibited potential antioxidant activity and inhibitory effect on AR.
