摘 要:采用响应面法对以木糖为碳源,土曲霉ASP-046菌株为菌种发酵衣康酸的培养基进行优化。在单因素试验的基础上,选取对衣康酸产量影响较大的培养基成份玉米浆、硝酸铵、磷酸二氢钾浓度进行Box-Behnken试验。结果表明,玉米浆对衣康酸产量的影响极其显著,其次是硝酸铵、磷酸二氢钾。最佳培养基组成为:玉米浆1.17 g/L、硝酸铵3.03 g/L、磷酸二氢钾0.06 g/L。在此条件下,衣康酸产量理论值为59.24 g/L,实际验证值为58.96 g/L,比优化前提高了11.9%。Response surface method was used to optimize the composition of medium for fermentation of itaconic acid with xylose as carbon source and Aspergillus terreus ASP-046 strain.On the basis of single factor experiment,the box-behnken experiment was carried out by selecting the main factors influencing the yield of itaconic acid in the medium,namely the concentration of corn steep liquor,ammonium nitrate and potassium dihydrogen phosphate.The results showed that the effect of corn steep liquor on the yield of itaconic acid was very significant,followed by ammonium nitrate and potassium dihydrogen phosphate.The optimum medium composition was corn steep liquor 1.17 g/L,ammonium nitrate 3.03 g/L,potassium dihydrogen phosphate.0.06 g/L.Under the optimal conditions,the theoretical yield of itaconic acid was 59.24 g/L,but experitmental was 58.96 g/L that increasing 11.9%higher than before optilization.
