摘 要:采用KOH-乙醇皂化再通过盐酸酸化的方法制备华山松籽油混合脂肪酸,利用硝酸银-硅胶柱色谱分离纯化华山松籽油中亚油酸,对纯化过程中硝酸银-硅胶对多不饱和脂肪酸(亚油酸)的动态吸附特性进行初步研究。在12.0 g活化的银化硅胶色谱柱中,加入与吸附剂比例1︰15脂肪酸,洗脱剂流速0.8 m L/min,以丙酮-正己烷为洗脱剂,可使华山松籽油中亚油酸由原来的57.7%柱层析分离后提高到98.12%。结果表明,以硝酸银-硅胶为吸附剂,采用柱层析法是一种高效分离亚油酸的方法。The fatty acids of Pinus armandi Franch seed oil were prepared by saponification with potassium hydroxide and ethanol followed by acidification with hydrochloric acid.The linoleic acid from Pinus armandi Franch seed oil was separated and purified by silver nitrate-silica gel column chromatography.The dynamic adsorption characteristic of polyunsaturated fatty acid by silver nitrate-silica gel in the purification process was studied.The 12.0 g silver-silica gel was in the chromatographic column,the fatty acids added and the ratio of the fatty acids to the adsorbent was 1︰15.Eluant flow rate 0.8 m L/min and acetone and normal hexane as eluent,after column chromatography separation,the purity of linoleic acid in Pinus armandi Franch seed oil increased from 57.7%to 98.12%.The results showed that the silver nitrate-silica gel as adsorbent,using column chromatography was a kind of high efficient separation method of linoleic acid.
