摘  要:目的探讨钙粒蛋白B(S100A9)在乳腺癌组织中表达及表皮生长因子(EGF)的反馈调节作用。方法收集在河北省唐山市开滦总医院行手术治疗的48例乳腺癌患者的病灶组织及癌旁正常乳腺组织,逆转录PCR(RT-PCR)和Western blot检测S100A9 mRNA和蛋白表达水平。体外培养人乳腺癌细胞系BT474,检测EGF处理12 h后及丝裂原活化蛋白激酶(MAPK)抑制剂PD98059处理2 h后细胞中S100A9、磷酸化细胞外信号调节激酶(p-ERK)mRNA和蛋白表达水平;使用siRNA-S100A9干扰序列下调细胞中S100A9表达,检测S100A9、p-ERK mRNA和蛋白表达水平。结果乳腺癌组织中S100A9 mRNA和蛋白相对表达量明显高于正常乳腺组织(P<0.01);乳腺癌组织中S100A9 mRNA高表达组乳腺癌患者病情复发率明显高于S100A9 mRNA低表达组(P<0.05)。经EGF处理后12 h后,10 ng/mL EGF组、1 ng/mL EGF组p-ERK、S100A9 mRNA和蛋白相对表达量明显高于空白对照组,10 ng/mL EGF组p-ERK、S100A9 mRNA和蛋白相对表达量明显高于1 ng/mL EGF组,差异均有统计学意义(P<0.05)。使用MAPK抑制剂处理BT474细胞2 h后,10 ng/mL EGF组p-ERK、S100A9 mRNA和蛋白相对表达量明显高于10 ng/mL EGF+10μmol/L PD98059组、空白对照组,差异均有统计学意义(P<0.01)。siRNA-S100A9组S100A9、p-ERK mRNA和蛋白相对表达量均明显低于空白对照组和siRNA对照组(P<0.01)。结论EGF可能是通过激活p-ERK可上调乳腺癌组织中S100A9表达。Objective To investigate the expression of S100A9 in breast cancer tissue and the feedback regulation effect of epidermal growth factor(EGF).Methods The lesion tissue and paracancerous normal tissue in 48 patients with breast cancer treated in the Tangshan Municipal Kailuan General Hospital were collected.The reverse transcription PCR(RT-PCR)and Western blot were used to detect the levels of S100A9 mRNA and protein.The human breast cancer cell line BT474 was cultured in vitro.The mRNA and protein expression levels of S100A9 and phosphorylated extracellular signal regulated kinase(p-ERK)in human breast cancer cell line BT474 were detected after EGF treatment for 12 h and MAPK inhibitor PD98059 treatment for 2 h;S100A9 expression was down regulated by using the siRNA-S100A9 interference sequence,and the mRNA and protein expression levels of S100A9 and p-ERK were detected.Results The relative expression levels of S100A9 mRNA and protein in breast cancer tissue were significantly higher than those in normal breast tissue(P<0.01);the relapse rate of the patients with breast cancer in the high S100A9 mRNA expression group was significantly higher than that in the S100A9 mRNA low expression group(P<0.05).After 12 h of EGF treatment,the relative expression levels of p-ERK,S100A9 mRNA and protein in the 10 ng/mL EGF group and 1 ng/mL EGF group were significantly higher than those in the blank control group,and the relative expression levels of p-ERK,S100A9 mRNA and protein in the 10 ng/mL EGF group were significantly higher than those in the 1 ng/mL EGF group,and the differences were statistically significant(P<0.05).After being treated with MAPK inhibitor for 2 h,the relative expression levels of p-ERK and S100A9 mRNA and protein in the 10 ng/mL EGF group were significantly higher than those in the 10 ng/mL EGF+10μmol/L PD98059 group and blank control group,and the differences were statistically significant(P<0.01).The relative expression levels of S100A9,p-ERK mRNA and protein in the siRNA-S100A9 group were significant
