摘 要:采用尼罗红染色和共聚焦激光扫描显微镜技术相结合,探讨有光处理、无光处理、室内阴干和自然晒干4种不同处理方式对采后油茶果油体及含油率的影响,并对制片过程中油茶果的切片厚度及染色时间进行优化。结果表明:切片厚度为60μm,染色12 h后,油体的观察效果较佳,油体均呈规则的圆形球状,直径分别为(23.97±8.94)、(10.57±8.01)、(10.93±6.51)、(16.16±1.34)μm;4种处理方式的油茶果仁含油率分别为53.08%、51.24%、48.95%、47.34%,与含油体率呈线性正相关,拟合度较高。Nile red staining and laser scanning confocal microscope(LSCM)were used to study the effects of four different treatments on the oil body and oil content of Camellia oleifera seeds,included light treatment,no light treatment,indoor shade drying and natural sun drying.The thickness and dyeing time of the slices were optimized.The results showed that the thickness of the slices was 60μm,and after dyeing for 12 h,the observation effect of the oil bodies was better.The oil bodies were regular and round,with diameters of(23.97±8.94),(10.57±8.01),(10.93±6.51)μm and(16.16±1.34)μm,respectively.The oil content of the four treatments was 53.08%,51.24%,48.95%and 47.34%,which was positively correlated with the oil content and had a high fitting degree.
