鸡源肠道乳酸菌的筛选及抑菌作用分析

摘　　要：为分离筛选耐酸及抑菌的乳酸菌菌株研制禽用微生态制剂,取健康鸡肠道内容物,利用MRS预还原厌氧琼脂平板培养基,对其乳酸菌进行分离培养,根据菌体染色镜检初步筛选出31株细菌,生化试验和16S rRNA序列比对分析鉴定皆为乳酸杆菌,其中包括7株罗伊氏乳杆菌、6株果囊乳杆菌、6株卷曲乳杆菌、3株口乳杆菌、2株唾液乳杆菌、1株短乳杆菌、1株母鸡乳杆菌、1株约氏乳杆菌、1株北里乳杆菌、1株桥乳杆菌、1株戊糖乳杆菌和1株Lactobacillus gigeriorum。耐酸试验中,有15株在pH值2.5的酸性环境下存活。挑选耐酸性强的7株菌进行抑菌试验,结果表明,7株菌对革兰氏阳性及阴性细菌都有一定的抑菌活性,且部分菌株混合抑菌效果比单一菌株抑菌效果好。In order to isolate Lactobacillus for preparation of probiotics used in poultry cultivation,the Lactobacillus isolates from chicken intestines was screened for strains with acid resistance and antibacterial capability.31 strains of Lactobacilli were isolated and cultured under anaerobic condition using pre-reduced MRS agar plate.The results of biochemical test and 16S rRNA sequence analysis showed that these Lactobacilli included seven strains of Lactobacillus reuteri,six strsins of Lactobacillus ingluviei,six strsins of Lactobacillus crispatus,three strsins of Lactobacillus oris,two strsins of Lactobacillus salivarius,one strain of Lactobacillus brevis,one strain of Lactobacillus gallinarum,one strain of Lactobacillus johnsonii,one strain of Lactobacillus kitasatonis,one strain of Lactobacillus pontis,one strain of Lactobacillus pentosus and one strain of Lactobacillus gigeriorum.In the acid resistance test,fifteen strains survived through the acidic condition of pH value 2.5.Seven of the fifteen strains with strong acid resistance were selected for the bacteriostatic test.The results showed that all of seven strains had certain antibacterial activity to both gram-positive and gram-negative bacteria,and the inhibition effect of some mixed was better than that of a single strain.
