摘  要:文章研究苹果多酚-壳寡糖微胶囊(APCM)对变异链球菌的抑菌及致龋效果。将未熟苹果多酚(APP)与壳寡糖(COS)按照2.5:1(w/w)的比例溶解后用喷雾干燥法制成APCM,同时将APP和COS作为对照,配制相应的浓度梯度后用纸片扩散法测定抑菌作用;微量肉汤稀释法测定MIC;分别观察对变异链球菌的生长抑制及产酸能力、生物薄膜形成能力、葡萄糖基转移酶(GTase)活性抑制、菌体黏附能力的影响。结果显示,APCM的MIC值为5 mg/mL,分别是APP和COS的50%和25%。APCM对变异链球菌生长的抑制作用是APP的2倍,COS的4倍;APCM在1.0 mg/mL浓度下,其产酸抑制效果接近于对照品0.1%NaF,在0.5 mg/mL的水平接近于COS和AAP在8 mg/mL时的产酸抑制效果;APCM的生物薄膜形成抑制效果在1 mg/mL是COS的8倍,在2 mg/mL是APP的4倍;APCM对GTase活性的抑制能力在2 mg/mL时高于COS在8 mg/mL的效果,在4 mg/mL时,高于APP在8 mg/mL的效果;菌体黏附拮抗效应显示,APCM在2 mg/mL时,已经超过APP和COS在8 mg/mL时的黏附拮抗效果。综上结果说明,相较于COS和APP,APCM对变异链球菌有更强的抑菌效果和抗龋活性。To evaluate the effect of apple polyphenols-chitooligosaccharides microcapsule(APCM)against Streptococcus mutans,which mainly lead to dental caries.APCM were prepared by spray-drying method with apple polyphenols(APP)and chitooligosaccharides(COS)ratio of 2.5:1(w/w).And then,Minimum inhibitory concentration(MIC),S.mutans inhibition activity,acid production inhibition activity,biofilm formation inhibition effect,glucosyltransferase activity(GTase)inhibition,and antagonism effect of bacterial adhesion were determined in vitro.Results indicated that MIC of APCM was 5 mg/mL,which about 50%and 25%of APP and COS level,respectively.The results of S.mutans growth inhibition of APCM was twice as much as APP and 4 times as COS.The inhibition activity of acid production showed that 1.0 mg/mL APCM inhibition effect was close to the control level of 0.1%NaF,while 8 mg/mL of COS and AAP reached that of 0.5 mg/mL APCM level,respectively.The inhibition effect of APCM on biofilm formation was 8 times as COS and twice as APP at the concentration of 1 mg/mL and 2 mg/mL,respectively.GTase inhibition results showed that 8 mg/mL of COS and APP reached that of 2 mg/mL and 4 mg/mL APCM level,respectively.The antagonism effect of bacterial adhesion showed that 8 mg/mL of COS and APP reached that of 2 mg/mL APCM level,respectively.All the above results show that compared with COS and APP,APCM had stronger antibacterial effect and anti-caries activity against S.mutans.
