摘 要:目的探究白藜芦醇抗人胰腺癌BXPC-3细胞增殖及机制.方法采用MTT法评价体外抗人胰腺癌BXPC-3细胞和人正常肝细胞LO2增殖活性,用流式细胞技术检测对BXPC-3细胞周期分布和凋亡的影响;利用紫外光谱、测定CT-DNA粘度等手段研究白藜芦醇与CT-DNA的相互作用;利用分子对接的方法模拟白藜芦醇与CT-DNA相互作用的模式.结果与结论白藜芦醇可与DNA发生相互作用,阻滞BXPC-3细胞的增殖于G0/G1期,诱导BXPC-3细胞凋亡.Objective To explore the effect of resveratrol on the proliferation of human pancreatic cancer BXPC-3 tells and its mechanism.Methods The resistance activity to the proliferation of human panrreatir cancer BXPC-3 cells and human normal hepatocytes L02 cells in vitro was evaluated through the MTT method.And the Flow cytometry was adapted to detect the effect on cycle distribution and apoptosis of human panrreatir cancer BXPC-3 tumor cells.The interaction with CT-DNA was researched by means of UV spectroscopy and determination of CT-DNA viscosity.The method of molecular docking was used to simulate the patterns of interaction with CT-DNA.Results and Conclusion Resveratrol can interact with DNA,block the proliferation of BXPC-3 cells in the GO/Gl phase and induce the apoptosis of BXPC-3 cells.
