摘  要:以单因素试验和正交试验研究超声温度、超声时间、料液比和乙醇浓度4个方面对荸荠皮中总黄酮提取率的影响.以Vc作为对照,通过·OH自由基、DPPH自由基、O2·自由基清除能力测试,研究其抗氧化活性.结果发现超声温度为60℃,超声时间40 min,料液比1:35(g/mL),乙醇浓度50%时总黄酮的提取率最大,为2.38%.荸荠皮总黄酮对·OH、DPPH、O2·自由基清除率与浓度呈正相关,当荸荠皮总黄酮浓度为600μg/mL时,·OH和DPPH自由基清除率分别为87.35%和93.45%,浓度为400μg/mL时,O2·自由基清除率为91.22%.试验结果认为超声波辅助单因素试验和正交试验提取荸荠皮中总黄酮合理可行,且提取物具有较好的抗氧化活性.The effects of total flavonoids from Eleochairis toberosa peels on extraction yield were studied,ultrasonic extraction temperation、ultrasonic extraction time、material-liquid ratio and ethanol content were tested by single factor.Meanwhile,the antioxidant activities was measured by scavenging of hydrexy、DPPH and O2-·free radical,with Vc as the control.The result showed that the uitrasonic temperation was 60℃,uitrasonic time was 40 min,material-liquid ratio was 1∶35(g/mL)and ethanol content was 50%,the yield of total flavonoids was 2.38%.The total flavonoids of E.toberosa peels on·OH、DPPH and O2-·showed concentration dependant.when the E.toberosa peels flavones content was 600μg/mL,·OH、DPPH the clearnce rates were 87.35%and 93.45%,when in flavones content was 400μg/mL,O2-·the clearnce rates was 91.22%.The conclusion showed that the single factor and orthogonal test by ultrasonic extraction E.toberosa peels total flavonoids is reasonable and feasible and the extract exhibited strong antioxidant activities.
