摘 要:目的:研究太行菊黄酮对D-半乳糖致亚急性衰老模型小鼠的体内抗氧化作用。方法:采用D-半乳糖法构建衰老小鼠模型,灌胃处理小鼠太行菊黄酮(100、150、200 mg/kg bw),计算肝脏、脑、心脏及肾脏指数,测定小鼠肝脏、脑、心脏及肾脏组织细胞中T-AOC、SOD、GSH-Px、CAT、MDA和GSH含量。结果:与NC空白组比较,衰老模型M组小鼠脏器指数显著下降(P<0.05),脏器组织细胞中除MDA含量增加外,T-AOC、SOD、GSH-Px、CAT和GSH含量均有所下降,且大部分差异达到显著水平(P<0.05);与衰老模型M组小鼠比较,小鼠脏器指数在太行菊黄酮处理各剂量组均有所增高,脏器组织细胞中除MDA含量降低外,T-AOC、SOD、GSH-Px、CAT活力和GSH含量均有所提高,太行菊高剂量组效果最显著,在部分脏器组织细胞中的效果甚至显著超过Vc阳性对照组(P<0.05)。结论:太行菊黄酮能提高组织细胞抗氧化酶活性和保护还原性物质含量、降低脂质过氧化物含量,衰老小鼠组织细胞的抗氧化能力从而得到显著提升,减缓衰老进程。Objective:The experiment focused on the in vivo antioxidation effect of the flavonoids from Opisthopappus taihangensis(FOT)on D-galactose-induced subacute aging mice.Methods:The mode of mice aging was established by subcutaneous injection of D-galactose in the back of the neck.The mice were induced by intragastric administration with different doses(100,150,200 mg/kg bw)of FOT to calculate the index of the liver,brain,heart,kidney,and the content of T-AOC,SOD,GSH-Px,CAT,MDA and GSH.Results:Compared with the blank group,the body organ index of the model group decreased significantly(P<0.05),and the T-AOC,SOD,GSH-Px,CAT activity and GSH content in each organ tissue decreased,but the MDA content increased.Most of the differences were significant(P<0.05).Compared with the model group,the organ index of the mice in each dose group of FOT increased,and the T-AOC,SOD,GSH-Px,CAT activity and GSH content in the organs increased to some extent.the MDA content was reduced,and the high dose group showed the most effective.Conclusion:FOT can increase the antioxidant enzyme activity and protective reducing substance content of tissue cells and reduce content of lipid peroxide,so that the antioxidant capacity of tissue cells of aging mice is significantly improved,and the aging process is slowed down.
