基 金:菌核侧耳培育关键技术及其活性研究。
摘 要:探讨固态培养菌核侧耳产菌丝多糖的培养基优化。通过单因素试验和正交试验设计,以菌质多糖含量为评价指标,考察固态培养基质、外加碳源、氮源和无机盐四个因素对菌质多糖含量的影响。结果表明:竹粉与桑叶质量比为2︰1,葡萄糖用量为1%,酵母粉用量为0.25%,Zn^2+用量为0.05%,菌悬液接种量为10%,发酵基质初始含水率为60%,于28℃恒温培养20 d,每瓶培养料中菌丝生物量(干质量)可达155 mg,菌丝多糖含量达13.8 mg/g。The culture medium optimization of fungal substance polysaccharide from Pleurotus tuber-regium(Fr.)Sing cultured in solid-state fermentation was discussed.Through single factor study and orthogonal experiment design,the effects of solid-state culture medium,carbon source,nitrogen source and inorganic salt on the content of fungal substance polysaccharide had been studied with the content of polysaccharides as the evaluation index.The results had showed that the mass ratio of bamboo powder to mulberry leaf was 2︰1,glucose dosage was 1%,yeast powder dosage was 0.25%,Zn^2+dosage was 0.05%,inoculum size of bacterial suspension was 10%,initial moisture content of fermentation substrate was 60%,the mycelium biomass(dry weight)in each bottle of culture material could reach 155 mg and mycopolysaccharide content could reach 13.8 mg/g when cultured at 28℃for 20 d.This study had provided a good research basis for fungal substance polysaccharide from Pleurotus tuber-regium(Fr.)Sing cultured in solid-state fermentation.
