摘 要:用大孔树脂纯化云芝多糖,并进行体外抗氧化活性研究。通过对10种大孔树脂进行静态吸附考察,确定最佳纯化树脂,对大孔树脂的动态吸附进行考察,筛选出最佳上样及洗脱条件,就纯化后的多糖对DPPH、ABTS、羟自由基的清除作用进行考察。结果表明,AB-8型大孔树脂为最佳纯化树脂;最佳上样条件为:pH 4.0、上样浓度5.0mg/m L、上样速率2.0 BV/h。最佳洗脱条件为:乙醇体积分数50%、洗脱剂用量3.0 BV、洗脱流速2.0 BV/h;纯化后的云芝多糖的纯度由46.81%提高到81.24%,对3种自由基都有很好的清除作用,且随着多糖质量浓度增大而增强。工艺稳定可行,为大别山区云芝资源的合理开发利用提供科学依据。Polysaccharide of Coriolus versicolor was purified by macroporous resin and its antioxidant activity was studied in vitro.The static adsorption of ten macroporous resins was investigated to determine the best purification resin.Through the dynamic adsorption of macroporous resins,the best loading and elution condition was selected.The scavenging effect of purified polysaccharide on DPPH,ABTS and hydroxyl radicals was investigated.The results showed that AB-8 macroporous resin was the best purification resin,and the best condition was pH 4.0,the concentration of sample 5.0 mg/mL,and the loading rate of sample 2.0 BV/h.The best dynamic desorption condition was ethanol concentration of 50%,amount of eluent 3.0 BV,the flow rate 2.0 BV/h.The purity of polysaccharide increased from 46.81%to 81.24%.The polysaccharide had different scavenging effects on the 3 kinds of radicals,and the clearance rate enhanced with the increasing of concentration.The technology was stable and feasible,which provided scientific basis for the rational development and utilization of Coriolus versicolor resources in Dabie Mountain area.
